Description

Abstract
我們先前研究已證實抗菌肽可在細胞膜上造成暫時性 ”孔洞(pore)”。是一種廣譜性殺(抑)菌。其中有一個抗菌肽-表面素,surfactin既為抗菌肽,又為生物介面活性劑biosurfactant,先前試驗結果表明surfactin是一種乳化劑(emulsifier)與皮膚助滲劑(transdermal penetration enhancer)。surfactin可造成在水溶液途徑中延長增加分子的透皮累積。在本計畫目標是將水產生物抗菌肽應用於化妝保養品。為達成此一目標,本計畫為期三年:第一年計畫中進行;1)進行抗菌肽(例,表面素的高產突變株篩選,2)開發液體發酵與固態發酵量產方式,3)將表面素添加於保養化妝品中測定其作為防腐劑、乳化劑、起泡劑與皮膚助滲劑等效果。第二年計畫,1)將含有抗菌肽與表面素添加之化妝品進行毒理學試驗, 2)申請國際化妝品料證號(International nomenclature cosmetic ingredients; INCI);第三年計畫,將與臨床皮膚科醫師進行水產生物抗菌肽與表面素作為化妝品添加劑之臨床試驗。本創新型抗菌肽可作為天然防腐劑、乳化劑與皮膚助滲劑,其應用層面廣泛,包括:化妝保養品製藥業具有巨大商機。 The present proposal relates to novel antimicrobial peptides (AMPs), from tiger shrimp (Penaeus monodon; monodoncin), grouper and Bacillus sp. These AMPs can be overexpressed in a heterologous expression system, such as yeast, by molecular cloning techniques or by induction technique. It had been demonstrated a wide-range of bacteriostatic and bactericidal effects on gram (-) and gram (+) bacteria, as well as fungicidal activities. This novel natural antimicrobial peptide can be used as preservative of cosmetics. We had isolated a novel surfactin (C53 H 93N7O13) is a biosurfactant which is produced through fermentation and purification of high-yield mutant Bacillus strain. The results of our previous studies demonstrated that this novel surfactin not only has anti-bacterial, anti-fungal and anti-virus effects, but also having emulsification and trnasdermal penetration enhancer activities. The goal of present proposal is application of aquatic antimicrobial peptides in cosmeceutic products. To fulfill this objective, we have established following short-term goals; In the first year of present poject, we have established high yield AMP (i.e. surfactin) mutant strain screen technique; 2) established liquid fermentation and solid state fermentation technique to mass production surfactin, 3) the biosynthezied surfactin will mix with cosmetics and/or skin care products and skin cleanness, oil emulsification, and surface tension abilities will be analyzed. Since the AMP can produced temporary “pore”on cell surface, therefore, the ability of AMP or surfactin which served as transdermal penetration enhancers will be determined. At second fiscal year, we will carry out; 1) the toxicology tests of personal care cosmetic product with surfactin-added, 2) filing the application of international nomenclature ingredient licence; In the third fiscal year we will collaborate with clinical dermatologist to carry out the clinical trial of these AMP additives in cosmetics, skin care products, ointment or other medicines.
