Description

Abstract
"開發「 新型式螢光轉基因觀賞魚種及品系」與「魚類不孕控制技術」已成為現在基因轉 殖觀賞魚產業重點方向及技術。我們實驗室(海大龔紘毅)與高海大陳鳴泉老師實驗室前期共同 主持與芝林企業合作的產學合作計畫,已經開發完成新穎魚類肌肉啟動子(promoter)/增強子 (enhancer),及三種不同顏色台灣珊瑚螢光蛋白基因cDNAs (TcRFP, TcCFP, TcGFP),並證實其 顏色及強度在基因轉殖斑馬魚均具有觀賞價值。根據我們之前實驗發現Tol2轉位酶在神仙魚 (Pterophyllum scalare)受精卵無法順利進行轉位作用。因此本計畫擬以另一近年來與Tol2轉位子 齊名已在昆蟲及老鼠成功建立基因轉殖動物之轉位子”PiggyBac”進行神仙魚之顯微注射基因 轉殖,以加速基因轉殖螢光神仙魚之建立。因此本產學合作計畫目的: 即是以我們已開發完成 之新穎斑馬魚(鯉科)和吳郭魚(慈鯛科) CKMb基因肌肉專一性啟動子/增強子,分別表現紅色 (TcRFP)、青綠色(TcCFP)、綠色(TcGFP)三種不同顏色台灣珊瑚螢光蛋白,以PiggyBac及Tol2 轉位子加速建立自有專利之新型基因轉殖各種螢光神仙魚(Pterophyllum scalare)及廣受喜愛的 小型鯉科四間魚(Barbus tetrazona)成為新的螢光觀賞魚種。" "Development of “new transgenic fluorescent ornamental fish species and strains” and “fish infertility control technology” has become critical directions and techniques in transgenic fluorescent ornamental fish industry. In our previous Academic-industry cooperation project with Jy Lin Trading Co., Ltd., we had invented the novel muscle-specific enhancer/promoter from zebrafish (applied for patents) and Mozambique tilapia and three different colors Taiwan coral fluorescent proteins (TcRFP, TcCFP and TcGFP) emitting red, cyan and green fluorescence. These three Taiwan coral fluorescent proteins had been validated in their strength and color in muscle of transgenic zebrafish and can be applied in transgenic fluorescent ornamental fish. According to our preliminary data by Tol2 transposon system, we found Tol2 transposase could not perform transposition in the angelfish (Pterophyllum scalare) fertilized eggs due to unknown reasons. In this project, we plan to use another well-developed PiggyBac transposon which is workable in transgenesis of many insects and vertebrate mice to promote establishment of transgenic fluorescent angelfish. The purpose of this Academic-industry cooperation project is to enhance development of new transgenic fluorescent fish species including middle ornamental fish angelfish (Pterophyllum scalare) and popular small Cyprinidae ornamental fish albino tiger barb (Barbus tetrazona) which expressing various Taiwan coral fluorescent proteins driven by the novel fish muscle-specific enhancer/promoter by the usage of PiggyBac and Tol2 transposn system, respectively."
