"本研究計畫為「固態培養牛樟芝酒精萃取物微奈米粒對糖尿病雄性大鼠生殖保護作用」。探討牛樟芝酒精萃取物微奈米粒的生理保護作用，解決因糖尿病併發的發炎性傷害問題，對代謝症候群的罹患者群體極為重要。近年國內外針對糖尿病併發的生殖傷害議題愈趨重視，主持人曾以不同來源牛樟芝萃取物進行相關研究，針對其抗發炎、調解血糖及生殖保健功效多有經驗心得，若結合微奈米包覆強化其劑量效果，將有利於深化整體理論基礎與實務產業應用。本計畫擬分三年期間聚焦於動物口服實驗的組織器官、血液生化值及細胞基因表現活性的變化分析上，實驗材料採用固態培養牛樟芝酒精萃取物，並製備成氧化矽- 幾丁聚醣微奈米粒。功效評估研究方法以國際間主流的糖尿病誘發雄性大鼠生殖傷害為基本架構，並採用近期較受認同的高脂飲食搭配STZ 模式探討對第二型糖尿病的血糖調節功效。第一年: 製備固態培養牛樟芝酒精萃取物、製備成氧化矽-幾丁聚醣微奈米粒、物化特性與安定性分析、動物口服預備實驗之劑量分組設計。第二年: 固態培養牛樟芝酒精萃取物微奈米粒之動物口服實驗、不同劑量投與之功效反應檢討。第三年: 固態培養牛樟芝酒精萃取物奈米粒之最適劑量化、探討吸收分布、代謝路徑與代謝基因體表現研究。" "The research theme is -The Amelioration Effects of Nanoencapsulated Ethanol Extract of Mycelia of Antrodia cinnamomea on Male Reproductive Function in High-Fat Diet /Streptozotocin-Induced Diabetes. In order to solve the adverse effects caused by inflammatory damage of diabetes mellitus/ metabolic syndrome, we will discussed the protential biological functions of micro-nano particles encapsulated ethanol extract of mycelia of A. cinnamomea (EMAC). It is very important issue, especially on male infertility to those suffering population as well as the natalist. In recent years, domestic and international research on this subject became into emerging and important. In our study, A. cinnamomea extracts from different sources and a variety of nutraceutic ingredients, combined with micro-nano-coated strengthen its protective effect. Our finding showed that this is a safe and effective method for preparation and processing. During the three years of the project, we intends to focus on animal experiments via oral administration. Including tissues and organs, blood biochemistry values and cellular gene expression wil be conducted. The solid-state culture of A. cinnamomea ethanol extract are selected as the experimental materials, and the silicon oxide -chitosan nanoparticles are prepared. Those designs will facilitate the future development due to intelligence protection and meet the industrial regulations. For methods, the male reproductive damage induced in diabetic rats as a international mainstream model. First year: preparation of encapsulated EMAC micro-nano particles, including the physicochemical properties and stability will be also analysed. Preliminary experiments for the oral administrated dosage by animal grouping. Second year: Animal experiments for EMAC micro-nano particles. The different dosage response according with their effectiveness will be overall reviewed. Third year: Determination of the optimal dosage of encapsulated EMAC micro-nano particles. Both the uptaking/absorption/distribution/metabolic pathway and the genomic expressing performance wil be elucidated."
