本申請案為東海長期觀測與研究（四）整合型計畫中的子計畫七。由於矽藻在海洋中是支持捕食食物鍊與幫助有機碳沉降的重要成員，以及三峽大壩截流後已大幅減少對東海矽鹽的供應量，所以申請人擬利用未來三年探討東海中的矽藻在此種環境變遷的過程中如何受到含矽營養鹽的限制。所採用的方法是利用矽鹽傳輸基因的表現量作為矽藻缺矽的指標，首先以總基因體定序及聚合脢連鎖反應調查東海中矽限制指標基因的岐異度，然後設計定量聚合脢反應引子並測試引子的專一性，最後進行海上實測，瞭解缺矽的區域與季節分佈。為求正確的解釋量測結果，也將在實驗室與海上進行矽鹽操控培養，以訂定基因表現量的上下限。另外將測量矽藻數量及種類組成，還有營養鹽誘發之螢光變化，作為解釋結果的輔助數據，這種以各種技術相互支援的策略應能以更確定的方式顯示出矽藻的生理狀態。至於矽藻受氮與磷限制的程度，也將配合執行中的另一個計畫一併測量。本申請案不僅將分子生物的觀念和技術帶入海洋生態的研究領域，而且能配合其他子計畫共同瞭解東海生態系在環境變遷的衝擊下會如何作出反應。 This proposal is the branch project 7 in the integrated research project “Long-term observation and research of the East China Sea – IV” (LORECS IV). Since diatoms in the ocean support the grazing food chain and help the sinking of organic carbon, and since completion of the Three-Gorges Dam has greatly reduced the supply of silicon to the East China Sea, the proposed research plans to investigate to what extent diatoms are limited by silicon in this changing environment in a period of 3 years. For this purpose, the expression levels of silicon transporter gene (SIT) family will be used as an indicator of silicon deficiency in diatoms. First, metagenomic sequencing and polymerase chain reaction (PCR) will be used to evaluate the diversity of this gene family in the East China Sea. Based on the result, suitable primers will be designed for quantitative PCR, and their specificity will be tested. The last step will be the measurement at sea, which should generate information of the spatial and seasonal distributions of silicon deficiency in the East China Sea. For a correct interpretation of the results, laboratory and field silicon manipulation experiments will be perform to define the maximal and minimal expression levels of SIT genes. In addition, diatom abundance, species composition, and nutrient induced fluorescence transients will also be measured as auxiliary data. Such a strategy by combining independent measurements to detect a single phenomenon will reveal the physiological states of diatoms with more precision. As for the possibilities that diatoms in the East China Sea may also limited by nitrogen or phosphorus, the related measurements will be performed by another ongoing research project. If granted, this proposal will not only bring the ideas and techniques of molecular biology to the field of marine ecology, but also work collaboratively with other branch projects under LORECS IV to understand the responses of the East China Sea ecosystem under the impact of environmental changes.
