Description

Abstract
觀賞魚為一具高經濟價值及發展潛力的養殖物種。由於種類繁多且色彩豐富,已逐漸成為僅次於犬貓的全球第三大寵物市場,極適合於人力成本高漲、水資源有限的台灣發展。本計畫合作業者-芝林企業有限公司為台灣最大觀賞魚出口商,在神仙魚品系培育已有多年繁養殖經驗,近來已篩選特殊顏色與外觀之神仙魚品系,但在遺傳改良仍有其潛在需克服問題,包括紅身、紅頭、藍身、白身及腹鰭等表型性狀之穩定提升。為能有效解決神仙魚之選種問題,本研究計畫目標在於利用分子標記輔助選育技術,提供業者開發與生產具國際市場發展潛力之神仙魚。本計畫共分三年度執行,包括:(1)依據神仙魚系譜與形質資訊,利用次世代定序之轉錄組與基因型定序,建立神仙魚觀賞性狀相關之功能性SSR與SNP分子標記資料庫,分析體表顯色變異之基因表現及探討訊息傳遞機制;(2)統計觀賞性狀相關功能性分子標記之基因型資訊,開發可輔助神仙魚觀賞性狀選育之分子標記;(3)利用已開發核苷酸變異序列之功能性分子標記,輔助神仙魚家系選拔並加以驗證,以建立分子標記輔助神仙魚選拔育種之應用平台。研究成果兼具基礎研究與產業應用價值,未來可提供業者建構完整產銷履歷及品牌認證之科學化管理,為觀賞魚產業注入新元素。 The ornamental fish are aquaculture species with high economic value and great development potential. They are very suitable for development in Taiwan which is experiencing rising manpower costs and limited water resources at present. In addition to cats and dogs, ornamental fish have become the third most popular pets, due to the great variety of breeds and the various gorgeous colors. The co-operative industry of present project is Jy Lin Trading CO., LTD., which is largest exporter of ornamental fish in Taiwan. They had many years of experience in numerous breeding angelfish cultivated strains, and the special color and appearance strains of angelfish have been discovered in most recent years. However, the industry is still needed to overcome the potential problems of genetic improvement and increase the inherently stable of phenotypic characteristics, including red body, red head, blue body, white body, and pelvic fins. In order to effectively solve the problem of angelfish breeding, the goal of this research project is to use marker-assisted breeding technologies to provide the industry would develop and product strain with high economic value and great development potential of angelfish in international market. This project, which is scheduled to last 3 years, will include the following objectives: (1) According to the characteristics based on pedigree information, the next-generation sequencing (NGS) technology of transcriptome (RNA-seq) and genotype by sequencing (GBS) methods will be used to screen and develop the SSR and SNP markers within the ornamental traits-related functional genes, to analyze individual samples for variation on the basis of differences in gene expression levels and discussion the signaling transduction mechanism; (2) The statistic method will be used to calculate the genotype information of ornamental traits-related functional molecular markers, which could be used to assist selection and breeding of ornamental traits in angelfish; and (3) The screened and developed molecular markers that have functional nucleotide sequence variations will be used to facilitate the family selection of angelfish, and their effects on ornamental traits will be validated in breeding experiments. Our findings will provide a platform for marker-assisted selection and breeding for industrial applications. This study assesses the results of basic research for their value to industrial applications. It will provide the ornamental fish industry to build the scientific management of complete traceability and certification, and to create an entirely new approach to productivity and competitive advantage in the future.
