放射治療是將放射能量準確的送到細胞核中，藉由破壞細胞核內的DNA 鍵斷裂，使細胞因而失去再生功能。在癌症放射治療過程中，腫瘤細胞與正常細胞均接受同樣放射線劑量，在抑制癌細胞生長的同時，也導致正常細胞損傷。放射治療的副作用包括肺臟、腎臟、肝臟、皮膚等組織產生纖維化，通常輕者造成組織紅腫潰爛、肌肉組織無法彎曲、聲帶纖維化失去聲音….等，重者則可導致病患器官功能衰竭致死。有鑒於組織纖維化之初期具可逆性，因此如何開發具抑制放療引發纖維化之藻類多醣體為本計畫研究要點。計畫第一年將自具量產能力之紅藻(麒麟藻)、褐藻(海帶/馬尾藻)中萃取多醣體，依據其醣組成及分子量進行分組，篩選出具高度抑制放療引發纖維化之藻類多醣體。接續第二年以In Vitro 模式探討藻類多醣體於預防/修復放療引發纖維化之機轉，以評估藻類多醣體之最佳使用方式 (放療前/放療後使用)。第三年透過In Vivo 模式探討藻類多醣體對組織纖維化之影響，以評估藻類多醣體之生物活性反應及最佳給予方式 (管餵內服/塗抹外用)。期望透過本計畫開發出具減緩放療副作用、輔助放射治療之藻類天然物。 Radiation therapy is commonly applied to the cancerous tumor because of its ability to control cell growth. Ionizing radiation works by damaging the DNA of cancerous tissue leading to cellular death. During the radiation therapy, tumor cells and normal cells accept the same dosage from radiation, and that leads tumor cells death and normal cells injury. The side effects of radiation therapy, including lung, kidney, liver, skin fibrosis…etc. Generally, patients may experience swelling, ulceration, muscle tissue can’t bend, loss of sound, and severe side effect can result to organ failure. In the view of early phase of tissue fibrosis is reversible, the aim of this study is to evaluate the effect of seaweed polysaccharides for anti-fibrosis after radiation therapy. At the first year, the polysaccharides are extracted from red (Eucheuma sp.) and brown seaweed (Laminaria sp. and Sargassum sp.) and then hydrolysis to small molecular weight (MW). According to their sugar profile, monosaccharide composition and MW, the seaweed polysaccharide with high inhibition of radiation-induced fibrosis is screened out. The second year, the In Vitro model is used to evaluate the optimal use of seaweed polysaccharides for prevention or repair radiation-induced fibrosis, and the signal transduction and mechanism of anti-fibrosis will also be evaluated. In the third year, the In Vivo model is used to evaluate the optimal use of seaweed polysaccharides on gavage or painting treatment to anti radiation-induced fibrosis. It is hoped that this project will develop natural seaweed polysaccharide that can reduce radiation side effects and assist radiation therapy.
