dc.description.abstract本研究探討利用蔗糖梯度離心配合分光光度法,是否可以有效地應用於綠豆澱粉分支酶(SBE)的分離與篩選。先進行5-45%廣範圍蔗糖梯度離心,再縮小範圍到1-25%,分別比較其回收區分中蛋白質與活性的分佈。結果綠豆粗萃取物的蛋白質之分子量密度範圍與SBE的總活性分佈主要集中在糖度l-10%間,此範圍不會干擾分光光度法酶反應的測試。SBE的比活性分佈顯示出蔗糖梯度離心可以將粗萃取物中的SBE分開成三個不同的族群,有純化效果,並具有再現性;又以5-45%蔗糖梯度之34%和42.5%左右的糖度範圍,與1-25%蔗糖梯度之15-20%和24.5%左右糖度範圍的純化效果較佳,其純化倍率分別為38倍、43倍、91倍與32倍。然而,經SDS-PAGE電泳分析各區分液,其主要蛋白質的圖譜沒有顯著差異。總結,蔗糖梯度離心與分光光度法配合,可以成功地用於篩選綠豆粗萃取物中的SBE,雖然蛋白質分開的效果不明顯,但是卻可將SBE純化,並分開成三個族群,有初步分離的效果。en_US
