Description

Abstract
無螯下目中的龍蝦科及蟬蝦科蝦類,多數種類皆為高經濟價值的海鮮,除了有詳細的形態特徵分類研究,亦有從分子生物基因的角度來探討其不同分類層級的演化親緣關係。隨著生物條碼基因的廣泛應用,此兩科蝦類雖為重要的水產品,卻無相關研究探討其生物條碼基因的差異性,訂定出物種界定的範圍,以供日後分子生物鑑種之依據;而部份分類層級仍未詳究其演化親緣關係。本計畫擬以動物界常用的生物條碼基因,即是粒線體第一細胞色素氧化酶基因 (COI) 片段序列,並加入粒線體16S 核醣體RNA基因 (16S rRNA) 片段序列,做為龍蝦科及蟬蝦科蝦類的條碼基因,並訂定出其不同分類層級的差異範圍。本計畫亦擬加入完整粒線體序列、新開發的細胞核蛋白基因以及生物地理分佈資料,配合原本常使用的分生基因標誌,來探究龍蝦科與蟬蝦科蝦類中,尚未被檢視之分類階層的地理演化親緣關係。本研究亦將以現場採集和與國外博物館商借樣本的方式,獲得較新且種類數多的研究樣本。研究結果除可將龍蝦科與蟬蝦科蝦類的分類鑑種方式,提昇至有完整生物條碼基因的佐證,亦可使此兩科蝦類在不同分類層級的演化親緣關係探究,更趨於全面且完備。The Achelata (Crustacea: Decapoda) includes Palinuridae and Scyllaridae lobsters and most of them are economically important seafood. Besides with the detailed taxonomic researches by morphological characters, the molecular phylogenetic relationships and evolutions have been elucidated on different taxonomic levels of these two families. DNA barcoding gene has been widely applied in many molecular identifications, but there is no any research to detect the similarity on the DNA barcoding gene of Palinuridae and Scyllaridae lobsters that can be further ranged for the species delimitation. Meanwhile, some levels within Palinuridae and Scyllaridae are still not clarified their phylogenetic relationship yet. This study will utilize the segment of mitochondrial COI gene which is the common DNA barcoding gene in animals, with the partial sequence of the mitochondrial 16S rRNA gene, to be the DNA barcoding markers for Palinuridae and Scyllaridae lobsters that can be employed to range the similarities on different taxonomic levels. Except the previously usual molecular markers, the study will also include the complete mitogenomes, the new nuclear protein-coding genes and biogeographical information for elucidating the phylogenetic relationships and evolutionary patterns on some undetected groups of Palinuridae and Scyllaridae. Acquisitions of fresh samples via field collections and diversified samples though exchanges with foreign museums are required. The results of this study are expected not only to rise the species identifications until assisting with the complete molecular information, but also to explain the evolutionary phylogenetics comprehensively at different classification levels of Palinuridae and Scyllaridae lobsters.
