Description

Abstract
微小動物性浮游生物是nano級(鞭毛蟲)及pico級(細菌)的主要攝食者,其中又以少毛類纖毛蟲 為大宗。因此在海洋生態系裡扮演著將有機與碳能量由微生物食物網傳遞至高階傳統食物鏈的關鍵 性角色。儘管先前已針對臺灣東北沿岸海域之少毛類纖毛蟲進行種類鑑定之研究,但是少毛類纖毛 蟲的組成及種類、數量周年變化並不清楚。因此本計晝希望利用蛋白銀染色法及無酸Lugol’s固定法 進行種類的鑑定及計數,同時以分子生物技術方法,獲得各個種類的18S rDNA序列,以進行臺灣東 北沿岸海域少毛類纖毛蟲之組成與生態之研究。研究主要分為三個部分:1)藉蛋白銀染色法鑑定少毛 類纖毛蟲種類;2)以18S rDNA序列分析探討少毛類纖毛蟲之生物多樣性;及3)利用無酸Lugol’s 固定之樣本計數少毛類纖毛蟲周年數量變化。以上研究有助於了解不同種類之少毛類纖毛蟲在同一 副熱帶沿岸生態系海域之數量變化,進一步作為後續針對各優勢種於微生物食物網中與環境的關係 及各自扮演的角色。"Microeukaryotes, mostly composed by oligotrich ciliates, are the major grazer on nanoplankton and picoplankton, and play an important role on the trophic link within microbial food web. Although previous studies have investigated the identification of oligotrichs ciliates in the northeastern coastal Taiwan, it is rarely known about the diversity of oligotrich ciliates and their annual species composition. Therefore, the goal of this project is to apply molecular techniques to elucidate the composition of oligotrich ciliates in three directions, including 1) the identification of the oligotrich ciliates by protargol staining methods; 2) the diversity of the species by 18S rDNA sequences analysis; and 3) the dial variation of species and its abundance by non-acid Lugol’s fixation. This study will strengthen our understanding of the functional role of different oligotrich ciliates in the microbial food web in a subtropical coastal ecosystem. Furthermore, the impact of global warming and acidification on the oligotrich ciliates could be monitored by observing the rapid change in their species composition, which further affecting the structure of the microbial food web."
