Description

Abstract
本計畫是『副熱帶陸棚浮游生物食物網生態過程研究』整合研究計畫之子計畫。主要在探討寡毛亞綱纖毛蟲種類組成在東海時空分布上的變化過程。微小動物性浮游生物,以寡毛亞綱纖毛蟲為大宗,扮演著將有機碳能量由微生物食物網傳遞至高階傳統食物鏈的重要角色。寡毛亞綱纖毛蟲是一群歧異度極高的原生生物,同時其營養模式非常多樣,因此在討論碳循環或環境變遷時,研究寡毛亞綱纖毛蟲的群聚結構是非常重要且必要的課題。為了探討研究海域寡毛亞綱纖毛蟲數量變化及種類組成,本研究不僅於倒立式顯微鏡下觀察計數寡毛亞綱纖毛蟲,同時搭配單一細胞PCR法,在同一玻片中挑取所有單一寡毛亞綱纖毛蟲細胞,並置於一微小離心管裡,最後將取得之18S rDNA序列作為種類判定的結果。本研究藉此實驗方法探討東海寡毛亞綱纖毛蟲種類組成與時空上的關係。實驗過程中每個細胞均拍照且紀錄大小,作為外部形態特徵之依據。最後將所得18S rDNA序列搭配外部形態特徵,整理一個東海海域寡毛亞綱纖毛蟲種類組成之資料庫,以作為後續研究之背景資料。This project is a component of the cooperative research program entitled “Plankton Ecological Processes in Subtropical-shelf Ecosystems” (PEPSE). Microeukaryotes, mostly composed of oligotrich ciliates, play an important role in the trophic link from the microbial food web to the traditional food web. With the highly biodiversity and variable trophic strategies in oligotrich ciliates, it is a MUST TO DO in the research of oligotrich community structures when discussing the carbon cycles and/or environmental changes. In order to discuss the variation of the abundance and the community composition of the oligotrich ciliates, the oligotrich ciliates will be counted under an inverted microscope and each cells will be isolated into a single Eppendorf by the means of the single-cell polymerase chain reaction for extracting its 18S rDNA sequence. The relationship between the abundance/composition of the oligotrich ciliates and their spatial/temporal distributions in the East China Sea by the process of the experiment. We will take a photo for the cell morphological characters and cell dimensions will also be recorded. A database of the composition of oligotrich ciliates will be made up by combining the morphological classifications and 18S rDNA sequences, for the background data of the research in the future.
