Description

Abstract
"本計劃的目的為發展去氧核糖核酸(DNA)雜合檢測之實驗室晶片,此晶片包含細菌 之裂解(cell lysis),DNA 之淬取(extraction)及濃縮,以及DNA 之雜合反應。其中檢測 之探針(Probe)帶有螢光,可以檢測螢光來決定目標DNA 與探針是否互補。首先含有細 菌之夜體通過微流道結構之薄膜纖維,此纖維孔徑大小足以阻擋細菌,如此濃縮之細 菌以解離沖刷,即可把細胞膜破裂,而使其中之DNA、蛋白質等釋出於水溶液中。當 此水溶液流經矽珠時,在高濃度鹽條件下,DNA 得以吸附在矽珠上,而可以從液體中 把DNA 分離出來,更進一步的以低濃度鹽沖刷則可以將DNA 從矽珠上淬取至水溶液 而得到純化且濃縮之DNA,這些DNA 通過先前發展成功的DNA 雜合晶片,即檢測出 是否與探針雜合反應,而完成一個實驗室晶片檢測過程。" "The proposed project develops a lab-on-chip for DNA hybridization assays, including cell concentration, cell lyses, DNA extraction, and final DNA hybridization with a specific probe. The chip includes a microdevice able to concentrate bacteria from sample solutions by a membrane of which DNA, proteins and other derbies are released from the cells. The DNA, in turns, are attracted to silica beads which the purified DNA are obtained. These DNA are hybridized with a specific probe and detected fluorescently. The microfluidic chip performs the hybridization assay, including cell lyses, DNA extraction and hybridization."
