Description

Abstract
"本計畫目的為尼羅吳郭魚抗病良種選育及研究抗病基因之作用分子機制。擬將台灣 不同尼羅吳郭魚品系進行基因體再定序工作,與Broad 研究所做的尼羅吳郭魚基因組 序列比較,建立不同品系DNA 變異資料庫包含單一核苷酸多型性、微衛星、微核醣核 酸及功能性基因DNA 變異及轉錄體表現資料庫。重點發展與基因相關之第一型分子標 誌,以提供吳郭魚抗鏈球菌之抗病育種分子標誌輔助育種平台。並擬以吳郭魚短型颗 粒蛋白前体基因做為吳郭魚抗病育種之分子標誌,以尼羅吳郭魚純系種魚進行雜交建 立不同尼羅吳郭魚品系雜交家系,以篩選不同數目短型颗粒蛋白前体基因家系進行抗 病家系育種。建立多個尼羅吳郭魚家系後,再以鏈球菌感染選育不同抗病力之尼羅吳 郭魚家系。以轉錄組分析比較對鏈球菌抗性強及敏感之尼羅吳郭魚家系之差異性表現 基因可只出參與抗病之功能性基因。進一步將以基因轉殖或基因剔除以驗證重要抗病 基因之功能及研究其分子機制。在此兩岸合作計畫中兩岸共同發展水產魚類遺傳育種 平臺技術,臺灣海洋大學團隊以尼羅吳郭魚為標的魚種,中國水科院黃海所團隊以牙 鮃為標的魚種,進行魚類抗病基因分子機制研究及抗病良種選育。" "The purpose of this project is selective breeding for disease resistance of Nile tilapia and study of molecular mechanism of disease-resistant genes. The whole genome of different Nile tilapia strains in Taiwan will be re-sequencing to establish database of Nile tilapia DNA variations including SNPs, microsatellites, microRNAs and functional protein-coding genes. We will focus on the development of gene-associated Type I markers to support markerassisted selection of disease resistance in Nile tilapia against Streptococcus. Various Nile tilapia families will be established by breeding of different Nile tilapia strains for family selection of disease resistance with the copy numbers of short form progranulin genes as molecular markers,. After treatment by Streptococcus, resistant and susceptible Nile tilapia families will be selected. Functional genes involved in disease resistance can be discovered by comparison of differential gene expression between Streptococcus resistant and sensitive Nile tilapia families by transcriptomics analysis. Furthermore, candidate disease resistant genes of Nile tilapia can be verified and studied by transgenesis and gene knockout. In this cross-strait cooperation project, NTOU research team in Taiwan will focus on Nile tilapia and research team in Yellow Sea Fisheries research Institute of Chinese Academy of fishery Sciences will focus on Japanese flounder to study molecular mechanism of fish disease resistant genes and selective breeding for disease-resistance by co-development of platform technologies in fish genetics and MAS."
