Description

Abstract
本研究擬針對海水魚弧菌症疫苗作有效性研究.上年度之研究結果得知Vibrio alginolyticus之LPS及菌體添加Levamisole保護效果較佳, 然而在第五、六週後RPS下降, 其保護效果似嫌短暫.因此有必要再繼續探討追加疫苗接種對延長保護效果的研究.本年度擬以不同處理之不活化菌苗、混合V.alginolyticus之脂多醣體或Levamisole後, 先施以浸泡或注射腹腔方式處理石斑魚苗, 再於第四及八週分別以不活化菌苗混合魚油配成飼料投餵一星期進行追加, 另一追加處理為注射腹腔方式進行.V.alginolyticus細胞外產物經不活化後與脂多醣體混合後, 以腹腔注射方式處理石斑魚苗, 再於第四及第八週追加不活菌苗如前試驗組, 另一追加處理同以注射腹腔方式進行.經以上處理後分別於第四、八、十二週後, 從尾柄處抽血和刮取魚體表黏液分析其抑制菌效果, 並同時以2倍的LD?菌液攻擊試驗探討其活存率, 以進行評估上述各試驗處理疫苗效果, 作為弧菌防治之參考. The study intends to investigate the efficacy of vaccines on vibriosis in cultured marine fish.In our previous studies, treatments using LPS from Vibrio alginolyticus and the bacteria supplemented with levamisole exhibited better protection in fish while their RPS decreased after 5and 6weeks.The duration of protection seemed to be short.Therefore, booster treatment studies to increase or enhance protection are needed.Various treatments of inactivated bacteria of V.alginolyticus mixed with LPS or levamisole will be used in this year's study.Juvenile groupers will be immersed or I.P.injected with tested vaccine prior to oral booster mixed with fish oil at week 4or week 8.The feeding will be tested for 1week.Another booster will be conducted by I.P.injection.Inhibitory activities against Vibrio alginolyticus in fish blood and mucus at week 4, 8or 12post-vaccination will be determined.In addition, two fold of the LD50of V.alginolyticus will be used to challenge all the tested fish to evaluate the efficacy of the vaccine and its usefulness in the control of vibriosis.
