Description

Abstract
"由於我國珍貴的保育國寶魚-台灣櫻花鉤吻鮭目前的放流效果不明,因此需要發展一套更精準的調查技術來評估放流之成效,並能持續監控整個族群的遺傳變異狀況。建立台灣櫻花鉤吻鮭親子鑑定之技術,可以廣泛應用於每年的保育工作及各項後續的研究上。 為求對國寶魚的傷害減低至最小,因此採用魚體身上之魚鱗與脂鰭為抽取DNA之材料。在利用黑鯛為練習對象成功後,接著採用種原相近魚種(大西洋鮭魚、虹鱒)已發表的 primer 嘗試攫取台灣櫻花鉤吻鮭的microsatellite DNA。 本計畫的研究結果如下:1.建立從黑鯛和台灣櫻花鉤吻鮭微量組織中抽取基因組(Genomic)DNA之技術。2.找到2對primer可攫取到台灣櫻花鉤吻鮭基因組DNA中的microsatellite DNA。雖然大西洋鮭魚的兩對引子可攫取出台灣櫻花鉤吻鮭的微衛星DNA,但是尚需在更多(100尾)的台灣櫻花鉤吻鮭魚隻上試驗以確認其有辨別個別台灣櫻花鉤吻鮭魚隻的能力(Avise 1994;Takagi et al., 1997;Taniguchi 1999)。此外本實驗使用洋菜膠體進行電泳檢視攫取微衛星DNA片段,其解析度尚待提昇。今後應該嘗試使用解析度更高,例如蛋白質(acrylamide gel)或序列(sequencing gel)電泳的方法,以證實此兩對大西洋鮭魚的引子確實具有辨別個別台灣櫻花鉤吻鮭魚隻的能力(Morris et al.,1996;Takagi et al., 1997;Martinez et al.,2000)。其次單一對引子尚不足以達到鑑定台灣櫻花鉤吻鮭親子的關係,要達到99%的親子鑑定的程度,必須要有4到5對引子,也即其辨識率接近10–9~10-10,意即2個個體相同的機率是1億7500萬分之1(Takagi et al., 1997;Taniguchi 1999)。找尋更多能夠攫取到台灣櫻花鉤吻鮭微衛星DNA的引子是未來值得努力的方向。" "This research addresses the application of a new group of molecular markers, microsatellite loci, to monitor genetic diversity of the endanger Formosan landlocked salmon population. Genetic diversity and population subdivision structure will be estimated based on microsatellite DNA analysis. We have tried 10 published primers designed from rainbow trout (Oncorhynchus mykiss) and Atlantic salmon (Salmo salar) and found four failed to amplify Formosan landlocked salmon. More primers will be tested and their polymorphic will be examined to confirm that microsatellite markers isolated from one species of salmonid are likely to be also useful in genetic studies of other salmonid species. The importance of microsatellite genetic marker for design of management strategies of Formosan landlocked salmon stock enchancement is emphasized. "
