Description

Abstract
"黑鯛第一年與第二年為雄性,第三年部分魚將性轉變為雌性,且以含雌二醇 (estradiol,E2)飼料投餵4 個月會誘導成為雌性,停止投餵後,會導致變回雌性 (reversible sex change),因此0-1 歲全雄性黑鯛提供為探討性別分化之模式魚種。本 研究之目的以組織學及分子遺傳基因之層次,探討遺傳基因在黑鯛性別分化與性轉變 之分子機制。實驗期間為三年,分別進行下列之實驗: 1) 探討3 個月齡至一年齡期間雄性黑鯛性別分化相關重要基因(Dmrt 1,Sox 9,Sf 1, Dax 1 與Amh)之關係。 2) 以E2 誘導性轉變後,再變為雌性為模式(reversible sex change),探討對雄性(Dmrt 1,Sox 9,Sf 1,Dax 1,activin 與Amh)與雌性(Wnt 4,follistatin,FoxL 2,Fig α 與aromatase)相關重要基因的關係。 3) 以in situ hybridization 方法確定Dmrt1 與Amh 在雄性生殖腺分化所表現的位置。 4) 探討精巢組織對卵巢組織發育之影響關係。 5) 以精巢與卵巢進行組織培養(in vitro tissue culture)的技術,探討精巢與卵巢組織 的關係。 6) 瞭解aromatase 在雄性黑鯛性別分化的影響。 7) 以Dmrt 1 RNA interference 的技術,探討Dmrt 1 在黑鯛生殖腺發育與性轉變之角色。 8) 檢驗精巢Amh 是否對精巢與卵巢發育扮演重要的角色。" "Black porgy, Acanthopagrus schlegeli Beeker, a marine protandrous hermaphrodite, is functional male for the first 2 years of life but begin to sexually change to female after the third year. Estradiol could induce a reversible sex change in 1-year-old fish.This sex pattern provides an unique model to study the molecular mechanism of male sex differentiation and sex change in gonad. The objectives were to investigate the molecular mechanism of gonadal sex differentiation in protandrous black porgy in the relation to male and female sex-related genes. The experiments were included as follows: 1) The expression profiles of Dmrt1, Sox9, Sf1, Dax1, activin and Amh transcripts in gonad during male sex differentiation in black progy. 2) The localization of Dmrt1 and Amh gene in the differentiating gonad by in situ hybridization. 3) The expression profiles of male genes (Dmrt1, Sox9, Sf1, Dax1, activin and Amh trascripts) and female gene (Wnt4, follistatin, FoxL2, Fig α and aromatase) in the gonad of E2-induced (fed) females. 4) The expression profiles of male genes (Dmrt1, Sox9, Sf1, Dax1, activin and Amh transcripts) and female gene (Wnt4, follistatin, FoxL2, Fig α and aromatase) during reversible sex chage from E2-induced female to a male. 5) The cause effect of testicular tissue to the ovarian development in the bisexual gonad by applying the surgical approach to excise testicular tissue. 6) The interactive effects of testicular tissue to the ovarian development by in vitro culture system. 7) The effect of aromatase in the sex differentiation of male ganad. 8) The specific roles of Dmrt1 in the male sex differentiation by Dmrt1 RNA interference technique. 9) The specific effects of Amh in the development of testicular and ovarian tissue in the bisexual gonad."
