Description

Abstract
"臺灣鮭魚(Oncorhynchus formosanus)是冰河期孓遺在臺灣的陸封型鮭魚,也是唯一存活在亞熱帶之太 平洋鮭魚,更是臺灣瀕危的特有(獨有)生物,因此臺灣鮭魚的復育已經刻不容緩。儘管目前臺灣鮭魚 野外族群超過 2000 尾,但是所有的個體皆由 200 尾左右的族群所衍伸,基因庫有弱化的現象。因此, 如何建立家系避免近親自交是相當重要的課題。此外,相較於其他馬蘇麻哈魚可以降海迴游,根據 初步研究臺灣鮭魚已經無法降海(適應鹽度變化),顯示上個冰河期後(1 萬年)的陸封隔離,已經弱化 臺灣鮭魚適應鹽度變化的能力。所以本計畫擬從臺灣鮭魚全基因體定序解碼來建立家系分子標記, 幫助復育且評估臺灣鮭魚降海迴游之能力(基因功能弱化的原因)。另一方面,本計畫將進行生存在淺 海熱泉環境之臺灣特有龜山島熱泉烏龜怪方蟹初步基因解碼與轉錄體分析適應高濃度硫化氫之可能 基因調控路徑,並探討地理分布位置相對遙遠且洋流流向不具有連貫性的 3 種怪方蟹,在演化過程 中是否具有(或曾經)一個廣泛分布的中間物種,探討 3 者之間在遺傳演化的關聯性。" "Taiwan salmon is a landlocked salmon and is the only surviving of the subtropical Pacific salmon. It is unique endangered (exclusive) species in Taiwan, so the restoration of Taiwan salmon is desperately needed. Although wild Taiwan salmon populations over 2000, but all the individuals reproduced from the 200 fish, gene banks are narrowed. Therefore, how to establish genetic makers for population to avoid inbreeding is a very important issue. In addition, compared to other Masu salmon had an ability to adapt seawater, Taiwan salmon lost this ability. This showing 10 thousand years separation reduced the ability of salinity adaptation. Therefore, this project aims to establish genetic makers for population to help Taiwan salmon preservation by whole genomic sequencing. In addition, to understand the reason for reduced function of salinity adaptation. On the other hand, this project also focuses on genomic sequencing survey and transcriptome of turtle island hypothermal vent crab. We try to elucidate the genetic correlation and species migration/evolution between 3 different vent crabs. In the further, to study the adaptive mechanism when crab stay in high level H2S condition."
