Description

Abstract
" 將文獻中已知的Bacillus licheniformis幾丁質.基因,以PCR方式選殖出來,經由原核表現系統---pET-20b(+)載體取得具酵素活性的重組幾丁質.,以蛋白質工程方式處理此幾丁質.,得此酵素的衍生分子,發現其衍生分子的幾丁質.具有轉醣基功能,即能產生幾丁六糖的水解產物。進一步研究其分子的轉醣基部位後,以基因融合方式轉接到其他不具轉醣基活性的他種幾丁質.,測試此新分子的幾丁寡糖生產率,並找出其最適條件。若能得此雙功能的複合酵素,則將對此分子進行人工演化(Molecular Evolution),並將對其結構與功能的關係以X-光結晶繞射做深度剖析。 "
