Description

Abstract
本實驗室在先前的研究中發現斑馬魚的sim2基因在發育初期即在間腦,腦室,鰓弧 以及腸胃等處有相當專一性的表現。在本計畫的第一期實驗中,即探討了sim2在斑 馬魚頭骨發育的過程中所扮演的角色。在本期的計畫實驗中,我們主要在探討環境 荷爾蒙戴奧辛對於頭骨發育的影響,發現了戴奧辛會造成斑馬魚頭骨機型發育的情 況,這項研究結果稍早由Teraoka等人發表(Toxicol Sci. 65,192-199)。但是本實驗 室在研究過程中發現色素抑制劑phenylthiourea (PTU)會與戴奧辛產生擷抗作用,明 顯降低戴奧辛所誘發的cyp1a1基因的活性。這項實驗推翻了一般對於PTU的看法, 認為PTU對於斑馬魚胚胎不會產生影響的推論。在實驗中,我們發現PTU本身會誘 發較低量的cyp1a1基因的表現,而這項基因的調控是受到AHR所控制。這項研究將 於今年七月發表於Biochemical Pharmacology。本計畫第三期,我們將持續對sim2以 及arnt2基因對於腦神經與頭骨的發育進行持續的研究,預期將會有所突破。During embryogenesis, sim2 mRNA appears in the animal hemisphere as early as 3 hpf and is expressed in a widespread pattern throughout the epiblast at the 75% epiboly stage, During the segmentation stae, sim2 mRNA is prominently expressed in the primordium of the hindbrain and appears as a transverse stripe in the epithelial layers of the mid-diencephalic boundary (MDB). In sim2-knockdown embryo, the craniofacial skeleton development was disrupted. To reveal the mechanism of TCDD-mediated defect in fish embryo, we have investigated the head skeleton development in TCDD-treated embryo by Alcian blue stain. It appeared that the development of cranial skeleton was disturbed. In addition, we found phenylthiourea (PTU) blocked TCDD-mediated cyp1a1 transcription via AHR signaling pathway. PTU induced a mild level of cyp1a1 transcription which was eliminated in AHR knockdown embryo. These results will be published (Biochemical Pharmacology 68 (2004) 63–71).
