Description

Abstract
本計畫之目標是結合功能性奈米材料(小分子、醣類、DNA、分子適合體和抗體修飾之金屬、金屬氧化物和半導體等奈米(微奈米和團簇)粒子)與纖維薄膜或膠體薄後應用於光譜、電化學與質譜分析應用。首先將合成新穎發光(金和銀微奈米粒子和團簇)、電化學活性(碳奈米管或石墨烯氧化物)或仿生物催化活性(合金和金屬氧化物)之奈米材料,經適當分子辨識修飾後,結合纖維薄膜或膠體薄膜後開發一系列生物分析技術。藉由不同辨識分子、奈米材料與薄膜組成來調控與目標分子(DNA、RNA和蛋白質(腫瘤標記))的結合能力。我們將提出三維空間的辨識方法來增加與目標分子結合力與選擇性。這些多樣性功能性奈米薄膜將利用光譜(吸收、螢光和拉曼等)、電化學與質譜方法偵測環境毒物(如重金屬和農藥)、生物大分子(如DNA、RNA和蛋白質腫瘤標記物)與癌細胞和病原體等。 The first goal of this proposal is preparation of functional nanoparticles via modification of small molecules, polysaccharide, DNA, aptamer or antibody with metallic, metal oxide or semiconductor nanoparticles, nanodots or nanoclusters. Subsequently, these functional nanomaterials will be conjugated with cellulose-membranes or gel-membranes through simple physical adsorption. The as-prepared functional nanomembranes were used as nanosensors coupled with optical spectroscopy, electrochemical meter and mass spectroscopy. The preparation of luminescent gold and silver nanodots or nanoclusters, electroactive carbon nanotubes and graphenes, enzymatic alloys and metal oxide nanoparticles were included in this proposal. After functionalization of these nanomaterials and on conjugation with membranes, these nanomembranes will be used in the development of novel bioanalytical techniques for detection of environmental toxins (heavy metal ions and pesticides), biomacromolecules (oligonucleotides and tumor protein markers), cancer cells and pathogens.
