Description

Abstract
"近年來,SARS、禽流感、登革熱、腸病毒……等各種病毒性傳染病陸續造成人類生命的 恐慌後,如何開發有效地預防及治療「病毒性傳染病」的保健食品或藥物,已成了全世界人們 心中的期待。登革熱(Dengue fever, DF)為登革病毒(Dengue virus, DEN)藉由蚊子叮咬而傳播的 傳染疾病,而腸病毒71 型(enterovirus type 71, EV-71)好發於年齡層以五歲以下孩童居多,因此 登革病毒及腸病毒儼然已是台灣地區地方性流行病之ㄧ。目前登革病毒及腸病毒尚無藥物及疫 苗可預防,僅能進行支持性的治療,因此開發有效預防登革病毒及腸病毒感染的藥物是迫切的 議題。樟芝(Antrodia camphorata)又稱牛樟芝(niu-chang-chih),為一種特有之真菌,只寄生於腐 朽中空的牛樟樹(Cinnamomum kanehirai Hayata)上,樟芝是非常重要的中國藥用真菌擔子菌 (Basidiomycetes),可用於治療食物及藥物之中毒、腹痛、高血壓、肝癌等疾病。目前有潛力之 化合物如固醇類、三萜類已被分離及鑑定出其成分,而且從樟芝子實體及液態培養之菌絲體萃 取出之多醣體已證明具有抗B 型肝炎病毒(Anti-hepatitis B virus)之活性。因此本計畫主要開發 具有抗登革病毒與腸病毒71 型的樟芝發酵產品及萃取物。在第一年計畫中,我們利用實驗室 已建立的細胞篩選平台,從54 種的樟芝及樟芝發酵產物篩選出具有抗登革病毒與腸病毒71 型 的潛力多醣萃取物。希望在第二年計畫中,我們將利用實驗室已建立的病毒感染小鼠模式,來 驗證篩選出具有抗登革病毒與腸病毒71 型的樟芝及樟芝發酵產物的功效,並探討其抗病毒的 可能機制。" "In this century, the health of human will be challenged by the newly emerged infectious disease, such as SARS, Avian flu, dengue fever and enterovirus. Hence, there is an urgent need for new treatments against viral infectious diseases. Dengue viruses (DENs) are a group of mosquito-borne flaviviruses that cause a mild febrile illness, dengue fever (DF), and infrequently cause a much more severe disease, dengue hemorrhagic fever/dengue shock syndrome (DHF/DSS). The enterovirus type 71 (EV-71) cause common enteric diseases in children, respectively, in summer of Taiwan. At present, the availability of safe and potent antiviral agents and vaccines against DENs and EV-71 is far from ideal. Antrodia camphorata (Chinese name, niu-chang-chih or chang-chih) is an exclusive fungus parasitic on the inner cavity of the endemic species Cinnamomum kanehirai Hayata and an important traditional Chinese medicinal fungus (Basidiomycetes) for the treatment of human diseases such as food and drug intoxication, diarrhea, abdominal pain, hypertension, itchy skin and liver cancer. Some bioactive compounds of A. camphorata including sesquiterpene lactone, steroids and triterpenoids have been isolated and characterized. Recently, polysaccharides extracted from fruiting bodies and mycelial cultures of A. camphorata have been shown to have anti-hepatitis B virus activities. In this proposal, we will try to separate, purify, and characterize the anti-DEN and EV-71 activity of fermentation products and extracts from A. camphorata by in vitro and in vivo model. In first year, we examine anti-DEN and EV-71 activity of potential polysaccharides from 54 kinds of fermentation products and extracts of A. camphorate by anti-viral screening cell model. Next year proposal, we will examine anti-DEN and EV-71 of potential polysaccharides from fermentation products and extracts of A. camphorate by DEN and EV-71 infected mice models and study the anti-viral mechanisms."
