Description

Abstract
"近年來,SARS、愛滋病、禽流感等各種病毒性傳染病陸續造成人類生命的恐慌後,如何開發有 效地預防及治療病毒性傳染病的保健食品或藥物,已成了全世界人們心中的期待。登革病毒(dengue virus, DENV)為一個具有醣蛋白外套膜的單股正向RNA 病毒,台灣在過去曾發生三次大流行,顯示登 革病毒感染在台灣為重要的流行性疾病。登革病毒主要藉由病媒蚊傳播感染至人體內,其感染後可能 會引起登革熱 (dengue fever, DF),或是更嚴重的疾病如:登革出血熱/登革休克症候群 (dengue hemorrhagic fever/ dengue shock syndrome, DHF/DSS)。然而免疫系統較差與第二次感染登革病毒的人 其可能更容易造成登革出血熱。由於登革病毒所引起臨床症狀的機制尚不明確,也無有效的登革病毒 疫苗來預防人類感染。然而近年來有越來越多的研究顯示出藻類硫酸多醣具有抗氧化、抗發炎、抗病 毒、抗癌、抗凝血等作用,為具潛力之天然食物。於第一年核定的計畫中,我們已經從台灣海域的褐 藻-半葉馬尾藻,以熱水萃取的方式製備出褐藻多醣萃取物,並且進行其多醣組成的分析,也利用利 用病毒斑抑制率試驗評估褐藻多醣萃取物抑制登革病毒吸附感染的效果。實驗結果如下:1. 褐藻多醣 萃取物中,半葉馬尾藻多醣硫酸根含量為29.2 ± 0.23%,而岩藻醣含量為79.96 g/g;2. 褐藻多醣萃 取物在病毒斑形成抑制率試驗中,以病毒感染細胞前加入抑制效果最好。因此,第二年計畫將延續前 述之結果,將利用本實驗室建立的登革病毒感染小鼠模式,來探討褐藻硫酸多醣萃取物中是否具有抑 制第二型登革病毒感染及免疫調節的能力,並進一步去探討其可能的作用機制及餵食褐藻硫酸多醣萃 取物是否可降低血球比容、減緩血小板低下與腸胃道出血情形,來提高感染小鼠之存活率。經由上述 的研究,我們希望能發展出有效的抗登革病毒及免疫調節的褐藻硫酸多醣萃取物,以作為開發抗登革 病毒及免疫調節的保健食品之應用。" "In this century, the health of human will be challenged by the newly emerged infectious disease, such as SARS, AIDS and Avian flu. Dengue virus (DENV) is a positive, single-stranded RNA virus with glycoprotein envelope, which caused three outbreaks in Taiwan. DENV infection has been shown an important epidemic disease in Taiwan. Dengue viral infections are one of the most important mosquito borne diseases in the world. They may give rise to dengue fever (DF), or a severe illness, dengue hemorrhagic fever (DHF) and dengue shock syndrome (DSS). People with weakened immune systems as well as those with a second or subsequent dengue infection are believed to be at greater risk for developing dengue hemorrhagic fever. Mechanisms of how DENV causes clinical symptoms are not fully understood. There is still lacking of effective vaccines to prevent human from DENV infection. Hence, there is an urgent need for new treatments against viral infectious diseases. Recently, a number of investigators have found that the extracts of algae present a broad range of biological activities including antivirus, antitumor, immunomodulation and anticoagulant activity. In the ongoing project, we prepared polysaccharide extracts from a brown algae (Srgassum hemiphyllum) around Taiwan sea area by using hot-water extraction methods and analyze their components of polysaccharides. We also performed in vitro experiments to evaluate the inhibition effect of polysaccharide extracts in viral adsorption by using plaque reduction assay on BHK-21 cells. The result showed that sulfated polysaccharides of Srgassum hemiphyllum were 29.2 ± 0.23%, and the fucose content of Srgassum hemiphyllum were 79.96 g/g. Data also showed that brown algae extracts has the most viral inhibitory effect when added before viral adsorption in the plaque reduction assay. The next aim of this proposal will use our laboratory previously established the dengue virus infected mice model to find out the anti-DENV and immuno-modulatory activity of brown seaweed sulfate polysaccharide extracts and investigate the possible mechanism. We study whether the brown seaweed sulfate polysaccharide extracts can attenuates hematocrit, thrombocytopenia and gastrointestinal bleeding, and improves survival rate in neonatal mice after dengue virus infection. Through all these efforts, we hope to develop that the brown seaweed sulfate polysaccharide extracts may be used as a health food supplement of anti-DENV and immuno-modulatory activity agents."
