Abstract
為了可以用作品管,確保產品質量,指標成分分析的建立,有助產品的標準化,及增加有效可信度。前期研究從台灣蜆的保肝確效及活性追蹤有效成分大量分離純化有效成分3,5,9-trihydroxy-cholest-7-en-6-one 和3,7,9-trihydroxy -cholest-5-ene,及topsentisterol D3都是膽固醇氧化物,所以本研究合成膽固醇相關的氧化類似物標準品。繼而建立分析系統,包括HPLC及LC-MS/MS分析方法的建立。此外,另一子項潘敏雄老師從台灣蜆中發現一個具抗肝纖維化的天然物,其結構具19個碳的cyclic acetal of glycerol。也建立分析系統以GC-MS分析。 The freshwater clams, clams, and oysters are popular seafood in Taiwan. Recently, the bioactive cholesterol oxidation products (COP) have been found in shellfish, the biological activities of COP including inducing apoptosis, antifungal, anticancer, and hepatoprotective activities. The aims of this study were to analyze the COP in the shellfish by LC-MS/MS and GC-MS. The four COP standards (3,5,9-trihydroxy-cholest-7-en-6-one, 3,7,9-trihydroxy- cholest-5-ene, cholesterol peroxide, and ergosterol peroxide) were used to analyzed Chanhung freshwater clam by thin layer chromatography and high performance liquid chromatography. To analyze the ergosterol peroxide, cholesterol peroxide, 7-hydroxycholesterol, 7-ketocholesterol, cholesterol-triol, 5,6-epoxycholesterol, 7-hydroxy--sitosterol, -sitosterol-5,6-epoxide in the shellfishes, COP were enriched by solid phase extraction, and subsequently analyzed by GC-MS, LC-ESI-MS/MS, and LC-APCI-MS/MS. Analyses of above 8 COPs in shellfishes by GC-MS were not succeessful, only cholesterol, -sitosterol, stigmasterol were detcted. On the other hand, no molecule ions peaks of COP could be detected by LC-ESI-MS/MS, all the of the COPs provided m/z 381.9, 437.9, 354.0. Nonetheless, whern LC-APCI-MS/MS was use to determine COPs in shellfishes, the results showed that only g/g COP in shellfish.
