Abstract
A. salmonmicida MAEF108與P. vesicularis MA103於含3 L基礎培養基發酵槽中以最佳培養條件發酵48 hr可得每mL 250活性單位(AU)與278 AU agarases。龍鬚菜寡醣濃縮液經Sephadex G-10膠濾層析確認析出液中寡醣類之分子量為724至1182 Da。以薄層色層分析確認可測得neoagarobiose, neoagarotetraose與neo-agarohexaose 等寡醣成分。25 L龍鬚菜寡醣水解液經濃縮與Sephadex G-10膠濾層析分離之析出液凍乾後,可得neoagarobiose, neoagarotetraose與neoagarohexaose之乾重量分別為7.25, 5.70及9.03 g。 The yields of A. salmonmicida MAEF108 and P. vesicularis MA103, while each cultivated in 3.0 L basal medium in a fermenter and under optimal incubation conditions, were 250 activity unit (AU)/mL and 278 AU/mL, respectively. The molecular weight ranges of concentrate of Gracilaria oligosaccharides solution were measured by gel permeation chromatography (GPC) and among 724-1182 Da. Thin layer chromatography was used to determine the contents of neoagarobiose, neoagarotetraose, and neo-agarohexaose while compared with opposite 2-6 sugar standards. The lyophilized powders of neoagarobiose, neoagarotetraose, and neo-agarohexaose, were weighed 7.25, 5.70, and 9.03 g, respectively, which were obtained from 25 L Gracilaria oligosaccharides solution via ultrafiltration concentration and Sephadex G-10 GPC.
