Description

Abstract
"菟葵毒(palytoxin; PTX)及河魨毒(tetrodotoxin; TTX)皆為臺灣常見的神經性海洋生物毒素, 近年來曾發生多起相關之水產食品中毒事件。本研究擬針對兩起因食用小沙丁魚及藍紋章魚引起之 PTX 與 TTX 中毒事件,檢驗其詳細毒成份與毒化物種來源,進一步自臺灣附近海域重新採集有毒物 種,分析毒成份並建立相關物種之基因資料庫以供後續研究參考。接著萃取臺灣常見之有毒 Palythoa 屬菟葵之 PTXs 毒素,進行包括溶血、酵素活性等各項毒性學測試,依毒作用所造成的生理活性特徵 建立可靠的檢測指標,並大量純化以 HPLC-UV與 LC-MS/MS 精準判別 PTXs 組成與濃度,標準化檢 驗操作流程,供未來相關單位中毒預防或中毒案例發生時之參考及應用。後續應用 LC-MS/MS 直接鑑 定海洋細菌生產之 TTX,採集有毒藍環章魚類物種探討其在不同季節下細菌相分布之情形,了解是否 具有河魨毒之生產能力,並以 LC-MS/MS 方法定性和定量,建立菌株生產 TTX 之最適條件,瞭解藍 紋章魚的致毒機制與季節分佈,期能大量純化以供後續研究利用。 " "Palytoxin (PTX) and tetrodotoxin (TTX) are both common neurotoxic marine toxins in Taiwan, which caused many poisoning accidents by ingesting seafoods in recent years. In this study, the toxicity and related species of sardine and blue-lined octopus which caused two poisoning cases will be investigated particularly. The related toxic species from the coast of Taiwan will be collected to analyze their toxicity and to establish the DNA data base for further comparison. Furthermore, the toxicity including hemolytic and enzymatic activity will be detected with PTXs extracted from Palythoa sp. in Taiwan, and the reliable standards are supposed to make according to the biological activity. The composition and concentration of purified PTXs will be also determined by HPLC-UV and LC-MS/MS analysis. In addition, the TTX-producing bacteria will be identified using LC-MS/MS. Different species of toxic blue-ringed octopus will be collected to detect the distribution of microflora in different seasons, and to realize the ability of producing toxins. The suitable conditions for producing toxins will be determined by detecting the quality and quantity with LC-MS/MS. The toxic mechanism and season distribution of blue-lined octopus are supposed to realize, and the toxins can be purified massively for further research. "
