Abstract
本研究為三年期長期計劃之第二 年,長程目標在界定那一種營養鹽會控制 超微浮游植物與大型浮游植物間的相對優 勢,並進而影響東海海域有機碳的流向及 食物鍊的結構。本年中雖曾進行培養實 驗,但主要工作為繼續前一年的工作,測 試流式細胞儀鑑別超微浮游植物的能力與 準確度,以便順利處理培養實驗的樣本。 我們以流式細胞儀計數東海及南海的水樣 後,發現目前的儀器設定可以清楚的辨識 聚球藻、原核綠藻、與微微真核藻等超微 浮游生物。另外在2002年5月22日基隆嶼測 站的水樣中測得超微浮游植物的垂直分 佈,其中聚球藻、原核綠藻、與微微真核 藻的最高細胞數分別為 6.5¥104、1.7¥104、 與 0.24¥104 cells ml-1。此為國內首次在東 海測得原核綠藻的豐富程度。This study is the second phase of a 3-year research project. The long term goal is to identify the critical nutrient species that controls the relative dominance between ultraphytoplankton and phytoplankton of larger sizes, and to examine how this change in dominancy affects the carbon cycling and food chain structure in the East China Sea. Although a number of nutrient enrichment experiments has been performed in this year, previous works on testing a newly purchased flow cytometer to evaluate its performance in identifying ultraphytoplankton and in cell counting precision were continued in this year. When the flow cytometer was used to analyze water samples from the South China Sea and the East China Sea, we found that the improved parameter settings could easily identify Synechococcus, Prochlorococcus, and picoeukaryotes based on cell size and pigment content. In addition, using samples collected at the Keelung Island Station on May 22, 2002, the vertical profiles of these ultraphytoplankton was established. The highest cell concentrations were 6.5¥104 , 1.7¥104 , and 0.24¥104 cells ml-1 for Syn-echococcus, Prochlorococcus, and pico-eukaryotes, respectively. This is the first time that the concentration of Prochlorococcus is measured in Taiwan
