Abstract
海洋生物毒中,最為人們所週知者為具神經性毒之河魨毒(Tetrodotoxin,TTX)和麻痺性貝毒(Paralytic shellfish poison,PSP)。由含有河魨毒之海洋動物中分離出可產生河魨毒之海洋細菌已被廣泛報告。為探討培養條件對細菌生產河魨毒能力之影響,將採自細紋玉螺體中具有河魨毒生產能力之Aeromonas hydrophila和V. alginolyticus為試驗菌株,在25.degree.C下分別將菌株培養於TSB (Trypticase soy broth)、YG(Yeast glucose) broth、NB(Nurtient broth)和BHI (Brain heart infusion) broth等培養基中,經振盪培養48小時後,以離心方式收集菌體。將菌體以超音波破碎部分純化後,以高效能液相層析儀鑑定河魨毒之含量。另以ORI(Ocean Research Institute) broth為基礎培養基,分別改變pH值、NaCl濃度、溫度及培養時間,以試驗菌株在四項條件下之河魨毒生產能力。結果得知A. hydrophila及V. alginolyticus在BHI培養基中之河魨毒生產能力較佳。V. alginolyticus之河魨毒生產性分別在pH8, 1%NaCl,25.degree.C,24小時有較佳之河魨毒生產量。A. hydrophila之河魨毒生產性分別在pH8,3%NaCl,30.degree.C,144小時有較佳之河魨毒生產量。 Among marine biotoxins, tetrodotoxin (TTX) and paralytic shellfish poison (PSP) are well known as sodium-blocking neurotoxins. TTX-producing bacteria had been found from many marine bacteria. To elucidate the optimum conditions for bacterial TTX production, the effect of various culture conditions on TTX formation was studied using one strain each of A. hydrophila and V. alginolyticus isolated from the lined moon snail N. lineata. The TTXs formation in these strains were proved by HPLC analysis. BHI (Brain Heart Infusion) broth gave a high toxin formation and the predominant toxin was identified as TTX in both A. hydrophila and V. alginolyticus. Optimum pH and NaCl concentration for both growth and toxin formation were pH 8 and 3% NaCl in A. hydrophila, respectively. But optimum NaCl concentrations for growth and toxin formation of V. alginolyticus were 3% and 1%, respectively. Various culture temperature and time also affected the TTX-producing capacity in both A. hydrophila and V. alginolyticus. These results suggest that various culture conditions influence the productivity of TTX production in these strains.
