Description

Abstract
分子檢測是具有高敏感度與高分辨度的診斷工具,隨著農漁業傳染性疾病的盛行,在水產養殖的現場對於高效率且操作簡單的分子診斷的需求愈來越高。我們認為多重聚合酶鏈鎖反應搭配毛細管電泳分析平台可以滿足水產養殖現場檢測需求,同時也是在效率與價格上最能符合市場的解決方案。本計畫擬針對蝦類養殖常見的四大傳染性疾病:急性肝胰臟壞死綜合症 (AHPNS)、微孢子蟲感染症,白點症以及十足目虹彩病毒感染症,開發出可實際在現場使用的高效率多重疾病檢測試劑,只要一個反應就能同時偵測這些疾病是否存在。在試劑開發上,擬採用高抗性 PCR 酵素、耐熱解旋酶、以及反應萃取液配方調整等技術,解決多重 PCR 對於現場樣品可能產生的反應性與專一性問題。透過本計畫的完成,除了可以有效降低水產疾病檢測的成本,實質幫助蝦類養殖漁民。未來也可以將本計畫所開發的技術搭配毛細管電泳平台應用在其他更廣大的分子檢測市場。 Molecular diagnostics are tools with high sensitivity and high resolution. Recently, more and more infectious diseases’ prevalence in agriculture and aquaculture, the demand for on-site molecular diagnostics, which is easy to use and high throughput, is increasing. We believe that the multiplex polymerase chain reaction (PCR) with capillary electrophoresis analysis platform can meet the demand, and is also the most cost-effective solution in the market. However, several technological breakthroughs in PCR reagents are necessary to improve the user experience and win over other competitors. This project is planned to apply several solutions for on-site multiplex PCR, such as PCR inhibitor-resistant polymerase; thermostable helicase, and customized reaction buffer system, to improve the detection specificity and sensitivity. Through the completion of this project, a commercial product that can simultaneously monitor common shrimp diseases, including Acute Hepatopancreatic Necrosis Syndrome, Enterocytozoon hepatopenaei, White Spot Syndrome Virus and Decapod iridescent virus, will be developed. In the future, the technology established by this project can also be applied to other molecular diagnostics.
