養殖吳郭魚的加工產業中，特別是台灣鯛冷凍魚片的加工，常伴隨而來大量的加工副產品如魚皮、魚頭及魚骨，一直是低度利用之加工素材；然而這些原料富含大量的蛋白質，如魚鱗、魚皮中的膠原蛋白及附著於魚頭及魚骨的肌纖維蛋白等。透過生物資訊工具的預測可得知，吳郭魚之膠原蛋白為多種活性胜肽(如血管張力素轉化酶抑制、抗氧化、抗健忘、調節血糖、抗血栓活性、調節免疫活性等)的先驅物，因此本計畫將著重於吳郭魚皮中膠原蛋白的利用，並藉由適當的萃取、濃縮、水解、純化及分析鑑定的手段可將這些低度利用的蛋白質轉化成高經濟附加價值的活性胜肽產品或基材，同時並探討其具有潛力的生理活性，以期應用於食品、化妝及生醫產業。本計畫預計以兩年的時間達成下列目標: 第一年: 1. 以淡水養殖吳郭魚皮為原料，進行樣品採集、前處理並製備膠原蛋白分離物及其明膠產物，並結合蛋白質體分析技術，鑑定其蛋白質結構及胺基酸序列的特徵。 2. 根據 27種常用蛋白脢，進行電腦篩選及預測最適合膠原蛋白及明膠的水解脢，以期生產具有特異性及不同效能之活性胜肽水解產物。第二年: 3. 執行 in vitro酵素水解及生理活性實驗，生產具有不同生理活性之膠原蛋白及明膠水解產物，並交叉驗證其生理活性的功能。 4. 進行膠原蛋白及明膠水解產物之體外消化及生理活性實驗調查，評估生產較適合人體消化吸收的活性胜肽前驅物。 Massive byproducts such as fish skin, heads and bones are produced from processing industrial of cultivated tilapias especially the processing of fillets from fresh water tilapias cultivated in Taiwan; generally these byproducts are underutilized processing materials which are relatively rich in proteins such as collagens from fish skin, and muscle proteins from residual flesh that are attached to fish bones and heads. According to the prediction by bioinformatics tools, collagens of tilapias contain precursors of multiple bioactive peptides (such as ACE inhibitor, antioxidant, antiamnestic, glucose uptake stimulating, antithrombotic, immunomodulating activities). Consequently, this project focuses on the utilization of tilapias collagens. With appropriate protein techniques of extraction, concentration, purification and identification, these proteins could be transformed to high value-added products of bioactive peptides or matrix applied in food, cosmetic and biomedical industrials. The following goals would be achieved in this two-year project: First year: 1. To conduct sample preparation, pretreatment and extraction of collagen isolates and gelatin products from skins of tilapias meanwhile using proteomic techniques to identify their protein structure and amino acid sequences. 2. To simulate the enzyme action on protein sequences with 27 common proteases and to perform computational screening of optimal hydrolyases for tilapias collagens gelatins prior to produce specific bioactive peptide products with different potential biological activities. Second year: 3. To execute enzyme hydrolysis and biological activities in vitro experiments in production of collagen and gelatin hydrolysates, and cross-check their actual biological activities. 4. To perform in vitro pepsin-pancreatin digestion with collagen and gelatin hydrolysates thereafter investigate their biological activities in assessment for precursors of bioactive peptides that are better suited for human digestion.
