By using a 15 WIG UV lamp and with a sperm-lamp distance of 6 cm, irradiation lasting for 2-4 minutes was tested to inactivate sperm effectively and to induce gynogenetic haploidy. Induction of gynogenetic diploidy, although low, can also be achieved by using irradiated sperm and heat shocking the eggs to 26°C at 25 minutes after fertilization for 25 minutes. In this experiment, we found that haploid syndrome as a diagnosis for haploid embryo was characterized by small eye. Satisfactory chromosome spreading was also obtained from young embryos of 14 to 20 days (under 12-15°C water temperature). |
