dc.description.abstract1972年12月至1973年2月,繼去年度之鳥魚精液保存試驗,再度配合東港分所之烏魚人工大量繁殖試驗,進行烏魚精液保存方法之探討。本次,首先研討烏魚精液之生物特性,續作5℃低溫及-196℃超低溫保存,略有所得: (1)在產卵洄游期游至東港沿岸之烏魚雄魚之GSI約為4.17~19.65不等,精液之pH值為7.4,精子之本體是2.3μ×1.4μ大小之長圓體,每cc精液平均約5.3×10^10個精子,精子在普通海水中具有最佳活力,鹽分較高或較低的水反抑制其活力。 (2)精液保存於5℃時,純精液至少維持活力15天,添加稀釋液之濃度各以Glycerin(1:1)5%,DMSO(1:1)5%,Sodium citrate 0.5%及1%,Glucose 3%,5%及6%以及Lactose 12%者較佳,但保存日數較不加任何添加物之純精液者減短。 (3)至於保存於-196℃液態氮之精液均添加稀釋液,目前已試驗部分係以添加Glycerin 5%及DMSO 5%之組最好,保存48天仍具活力,唯因其他處理仍在保存中,尚待比較方能定論。 (4)此次授精情形最好者為添加Glycerin 5%,保存於-196℃中39天之精液,其受精率及孵化率分別為15.63%及61.33%。en_US
