Abstract
台灣養殖石斑魚病毒疫情較其它魚種或地區更為複雜,除了發現有石斑魚神經壞死病毒感染之外,分屬蛙病毒屬和巨細胞病毒屬之兩種虹彩病毒都曾在台灣養殖石斑魚被分離出。我們先前的研究結果更顯示,兩屬虹彩病毒共感染會導致石斑魚出現不同的死亡模式,或許因為如此,長期來台灣石斑魚養殖場的病毒病疫情難以防治。然而,病原共感染對養殖魚類的影響至今研究仍相當有限,因此本計畫擬以石斑虹彩病毒 (grouper iridovirus, GIV)為主軸,探討與其他兩種病原病毒,神經壞死症病毒 (grouper nervous necrosis virus, GNNV) 和臺灣紅彩病毒 (grouper iridovirus of Taiwan, TGIV) 共感染在細胞 (in vitro) 以及活體內 (in vivo) 的交互作用,進而釐清病毒共感染在石斑魚造成致病性與拮抗作用。第一年,模擬疫情發生現況,分析先感染GNNV後感染GIV,以及GNNV/GIV共感染在細胞與活體的交互影響;另外探討兩屬虹彩病毒 (GIV/TGIV) 共感染在石斑魚體內的表現消長情形。第二年利用轉錄體學分析以上幾種共感染情形下魚體內之相關基因表現,並進行組織病理切片觀察,以期透過KEGG路徑和組織病變情形,釐清共感染對導致石斑魚死亡的可能交互作用機制。 The outbreak of viral diseases in grouper aquaculture in Taiwan is more complicated than other fish species or regions. In addition to the grouper nervous necrosis virus infection, two iridoviruses belonging to the genus Ranavirus and genus Megalocytivirus have been isolated simultaneously. Moreover, our previous research results show that the co-infection of these two iridoviruses can cause different mortality patterns of groupers. Perhaps because of this, it has been difficult to control the virus disease outbreak in Taiwan grouper farms for a long time. However, relevant researches about pathogen co-infection in aquatic animal are still less, so this project aims to investigate the coinfection of GIV with the other pathogenic virus, grouper nerve necrosis virus (GNNV) or grouper iridovirus of Taiwan (TGIV) in vitro and in vivo. Furthermore, clarify the pathogenicity and antagonistic relationship caused by viral co-infection. In the first year, simulate outbreak situation, GNNV will infect first and follow by GIV infection, besides GNNV/GIV co-infection will carry out and all of these combination will analyze in vitro and in vivo. Moreover, the fluctuation of GIV/TGIV expression in co-infected groupers will estimate. In the second year, transcriptome sequencing and histopathological observation of fish under the above-mentioned co-infection situations will evaluate in order to clarify the possible interaction mechanism of co-infection on the death of grouper through KEGG pathway and pathological changes.
