"珊瑚礁生態系具有豐富的生物多樣性，但因環境變遷及人為破壞，珊瑚數量逐漸減少，人工繁養殖珊瑚成為現在重要的議題，本研究以有性生殖、細胞生物、分子遺傳與分子內分泌的角度，期望建立一個先進的養殖系統，使得珊瑚能在人工環境下進行有性生殖及刺激配子排放，但目前關於珊瑚有性生殖的機制還有許多地方尚未瞭解，若找到與生殖腺發育、成熟及有性生殖機制相關之因子，對人工繁養殖珊瑚將有很大的幫助。本研究目的主要利用腎形真葉珊瑚 (Euphyllia ancora) 為實驗動物，以抑制性扣減雜交法、生殖腺轉錄體、原位雜合反應及其他分子生物與免疫化學組織染色，進行刺絲胞動物有性生殖、生殖細胞發育與成熟機制之研究。目前已有許多預備實驗成果支持此計畫，在三年期間將利用腎形真葉珊瑚與分子生物之技術，致力於探討及回答以下問題： (1)雄性珊瑚在繁殖季與非繁殖季哪些基因有表現上的差異或具有特異性？ (2)雌性珊瑚在繁殖季與非繁殖季哪些基因有表現上的差異或具有特異性？ (3)比較雄性及雌性珊瑚在繁殖季中哪些基因有表現上的差異或具有特異性？ (4)從繁殖季中有基因表現上差異或有特異性之基因，分析其在生殖腺中的表現位置(體細胞或生殖細胞或兩者皆有)？ (5)進一步分析該基因是否為賀爾蒙(成長因子、胜肽、類固醇或其他物質)？ (6)是否可以利用所找到的賀爾蒙或相關之化合物在人工養殖環境下刺激珊瑚的生殖腺發育或成熟？本研究極具學術創新性、重要性並具有高度的應用價值，為國際學術界珊瑚領域最前端之珊瑚生殖生理與有性生殖的研究，本研究計畫將對於海洋珊瑚礁生態環境保育具有重要的學術意義及應用價值。" "Coral reef ecosystems possess one of the highest biodiversities on earth. However, due to unfavorable environmental factors and human impacts, many coral reefs are declining in population number worldwide. Aquaculture of scleractinian corals is then expected as one of the excellent methodologies to propagate target coral species and conserve their genetic resources. Better understanding of the intrinsic factors controlling sexual reproduction, germ cell development and maturation in stony corals would allow us to establish an advanced system for propagating corals under aquaculture condition. So far, however, little is known about the molecular and endocrine mechanisms underlying sexual reproduction of stony corals. To address this issue, using a stony coral Euphyllia ancora with genetic, molecular and cellular approaches, we attempt to answer following questions in this 3 years project. (1) What are the differences in the gene expression patterns in female coral gonad between reproductive and non-reproductive season? (2) What are the differences in the gene expression patterns in male coral gonad between reproductive and non-reproductive season? (3) What are the differences in the gene expression patterns between female and male corals in reproductive season? (4) Which type of cells in the gonadal region somatic cells, germ cells, or both of them expressed the above identified genes? (5) Are there any genes regarding hormones (growth factors, peptides, steroids, and their receptors) in the gonadal transcriptome? (6) Can we induce the germ cell development and/or maturation under the aquaculture condition by hormone treatment? We have already started to study a part of this project and obtained some excellent preliminary data which could fully support our current project. The present project possesses very strong novelty, significance and will be a cutting edge in the field of reproductive biology of stony corals in the world."
