近年來，SARS、愛滋病、禽流感等各種新興病毒性傳染病陸續造成人類健康的威脅，如何開發有效地預防及治療病毒性傳染病的保健食品或藥物，已成了全世界人們心中的期待。流行性感冒病毒(Influenza virus) 所引起的急性呼吸道傳染病，與一般感冒不同，其症狀較為嚴重，此疾病重要性在於其爆發流行快速、散播範圍廣泛以及併發症嚴重。另一方面，流感可以同時侵犯所有的年齡層，因此開發有效預防流感病毒感染的方法是現今迫切的議題。近來的研究文獻指出薑黃素具有抗氧化、抗發炎、抗癌、抗病毒等功效，但有溶解性及生物可利用性差的缺點。近年來，奈米材料被廣泛應用於醫療用途，傳統的奈米材料是由重金屬元素所組成，具有毒性且生物相容性差，而作為碳奈米材料的新成員碳量子點容易接上官能基，使其具有更好的生物相容性和更低的毒性及低成本等優點。因此本計畫將利用細胞與動物模式，探討薑黃素及其碳量子點對於A型流感病毒(H1N1)感染之保護效果。首先，將薑黃素使用不同溫度以乾燒法合成碳量子點，並分析其性質。然後，在細胞實驗依病毒感染前、中、後加入薑黃素及其碳量子點，再以細胞存活率(MTS assay)、病毒斑試驗(plaque assay)、病毒RN表現量測定(real-time PCR)及病毒抗原蛋白分析(Western blotting)等實驗進行抑制效果評估，再分析薑黃素及其碳量子點抑制病毒感染之機制。最後，在BALB/c小鼠感染A型流感病毒模式中，探討薑黃素及其碳量子點抑制A型流感病毒感染的能力、抑制途徑及調節肺部發炎反應之作用機制。經由上述的研究，我們希望能發展出有效的抗A型流感病毒的薑黃素及其碳量子點，並瞭解其抗病毒機制，以作為抗A型流感病毒保健食品之應用。 In the past decades, human health has been challenged by the newly emerging infectious disease, such as SARS, AIDS, and avian flu and so on. There is an urgent need for new treatments against viral infectious diseases. Influenza is a viral infection that affects mainly the nose, throat, bronchi and lungs. This virus is transmitted easily from person to person via droplets and small particles produced when infected people cough or sneeze. Influenza tends to spread rapidly in seasonal epidemics. Influenza can lead to severe complications, including severe pneumonia and death. The cheap, easily available anti-influenza treatment with no or tolerable side-effect is urgently needed. Recently, curcumin have been shown broad biological activities, including antiviral, antitumor, immunomodulatory and anticoagulant activity. But poor solubility and bioavailability are their big drawbacks. In recent years, nanomaterials have been widely used in medical applications. Traditional nanomaterials are produced from heavy-metal elements, which suffers from poor biocompatibility and high toxicity. As a newcomer from the carbon nanomaterials family has been gathering considerable attention in the nanotechnology field due to their low toxicity, excellent biocompatibility, and low price. In this project, we will investigate the protective effects of curcumin and its carbon quantum dots on influenza A virus (H1N1) infection on in vitro and in vivo model. Firstly, we will use curcumin to synthesize carbon quantum dots by heating method at different temperatures, and analyze its properties. Then we will evaluate the cytotoxicity of curcumin and its carbon quantum dots by using cell viability assay (MTS assay). Curcumin and its carbon quantum dots will be added before, during, and after viral infection followed by plaque assay to investigate the anti-viral efficacy. We will utilize plaque reduction assay, viral RNA expression (real-time PCR) and viral antigens assay (Western blotting) to study the anti-viral effects of curcumin and its carbon quantum dots. We will investigate the preventive or therapeutic effects and immunomodulation mechanisms of lung induced by curcumin and its carbon quantum dots on the influenza virus A infected BALB/c mice model. Taken together, we will understand the antiviral effects and mechanisms of curcumin and its carbon quantum dots. Hopefully, the extracts of curcumin and its carbon quantum dots may be developed as a supplementary agent of anti-influenza virus A therapy.
