13株產agarase海洋細菌已完成初步鑑定,在初期測試條件下以Moraxella spp. Aga14、Flavobacterium spp. B13、Pseudomonas vesicularis MA103、Aeromonas salmonicida MAEF108 等四菌株之agarases產量較高。進行此四株海洋細菌生產agarase的最佳個別培養條件與培養基組成的實驗,所得結果顯示P. vesicularis MA103最高可得到 15.73 unit的agarase activity。綜合個別最佳反應條件進行培養時,A. salmonicida MAEF108可以得到71.35 unit 的agarases最高產量。P. vesicularis MA103所產agarases 經初步純化分離後,已可分離得到二個蛋白質,分別稱為agarase I 與agarase II。其中agarase I之分子量為42.7-50.3 kDa。而agarase II則仍待深入探討。酵素處理方法萃得之龍鬚菜海藻多醣以P. vesicularis MA103所產初步純化agarases 分解為寡多醣類混合物後,於動物細胞組織培養實驗中已呈現具有免疫活性之結果,但該部份之實驗仍待本年度延續計畫的工作中,以發酵槽大量生產適當的agarases以用來分解生產海藻寡多醣混合物後再繼續深入探討。 The primary identification of thirteen marine bacterial isolates had been complished. Four marine bacteria, Moraxella spp. Aga14, Flavobacterium spp. B13, Pseudomonas vesicularis MA103, and Aeromonas salmonicida MAEF108, showed higher agarase activities in their cultured broth than other nine marine agarase producing strains. After examining the individual incubation condition and the major components of the media for these four agarases producing strains, the results indicated that the highest agarase activity could be obtained was 15.73 unit by P. vesicularis MA103. While combining those optimal individual incubation factors together, A. salmonicida MAEF108 could increase its agarase yield up to 71.35 unit. The agarases that produced by P. vesicularis MA103 had been primarily purified, and two proteins were separated and then named agarase I and agarase II. The molecular weight of agarase I had been determined as 42.7-50.3 kDa. As to the determination of molecular weight of agarase II, there is more work to do. The algal poly-oligo-saccharides mixture, which derived from the enzyme-treated Gracilaria alagl polysaccharide extract digested by the agarase produced from P. vesicularis MA103, had been examined their immune activity with the animal cell tissue culture technique, and the preliminary results showed positive reaction on the immune activity increasing of those tested cell culture. But the final results could be accomplished only when more algal poly- and oligo-saccharides mixture could be obtained during this year's work by the production of agarases with fermenter.
