對蝦包括草蝦Penaeus monodon、斑節蝦Marsupenaeus japonicus及白蝦Litopenaeus vannamei為國內及世界其他大部分地區飼養的蝦種。然而此等蝦在養殖過程常遭受到弧菌如Vibrio harveyi 、V. alginolyticus等以及病毒如MBV (monodon baculovirus)、WSSV (white spot syndrome virus)等感染，造成嚴重經濟損失。已知疾病的爆發與衰弱宿主、不良環境以及病原入侵相互作用引起。環境突變及惡劣環境影響蝦的生理代謝反應，降低蝦的免疫力及抵抗弧菌能力，因此，在水產養殖中，除了維持良好的環境、提供適當的營養外，提昇蝦的免疫能力，避免病原感染為重要的課題。本計畫擬找出適當的銀奈米顆粒及濃度，控制養殖環境的弧菌，研究弧菌在銀奈米存在下的感受性，研究銀奈米影響弧菌的成長及死亡，研究銀奈米的抗菌特性，以及研究不同形式銀奈米顆粒與弧菌關連。 Penaeid shrimps like Penaeus monodon, Marsupenaeus japonicus, and Litopenaeus vannamei are the major commercial species currently being cultured in Taiwan and other regions in the world. However, shrimp farming has suffered serious problems linked to infectious disease due to vibriosis such as Vibrio harveyi and V. alginolyticus, and several species of viruses such as MBV (monodon baculovirus) and WSSV (white spot syndrome virus) resulting in vast economic losses. In an intensive culture system, shrimps are under the influence of changes in physico-chemical parameters like temperature, salinity, pH and low dissolved oxygen, and metabolites such as ammonia, nitrite, and sulfide. The interactions of three things: environment, host and pathogen are required for an occurrence of disease outbreak. It is known that deteriorated environment affect the metabolic physiological response, decrease the immunity of shrimp and its resistance against Vibrio alginolyticus. Therefore, increasing shrimp’s immunity, and prevention of pathogen infection are of primary concerning. The objective of the proposal is to formulate the concentration of silver nanoparticles to control the Vibrio population in the culture ponds. The proposal is 1) to determine the antimicrobial activity of silver nanoparticles to the pathogenic Vibrio isolated from the shrimp farm, 2) to examine the susceptibility of Vibrio spp. to different silver nanoparticles, 3) to examine the Vibrio spp cells growth or killing kinetics in the presence of silver nanoparticles, 4) to evaluate the shape dependence of the antimicrobial activity of silver nanoparticles against Vibrio spp, and 5) to study the silver nanoparticles of different shapes and their interactions with Vibrio spp.
