Keywords: | Ganoderma Iucidum CCRC 36124;Ganoderma lucidum CCRC 36124, Liquid culture, Mycelial growth, Polysaccharide formation;液態培養;菌絲生長;多醣生成 | Issue Date: | 1-Oct-2004 | Publisher: | 臺灣農業化學會&臺灣食品科學技術學會 | Journal Volume: | 42 | Journal Issue: | 5 | Start page/Pages: | 350-355 | Source: | Taiwan Journal of Agricultural Chemistry and Food Science | Abstract: | 本研究針對靈芝Ganotlerma lucidum CCRC 36124菌株於液態培養中之菌絲生長及其多醣體之生成,進行三角瓶振盪培養條件探討。在所測試的數種碳源中,以brown sugar和malt extract對菌絲生長及多醣生產效果較佳。yeast extract 和 skim milk為較佳之氮源。在油脂的添加方面,在培養基中加入紅花籽油可有效提升菌絲產量。再者,添加CaCO(下標3)對菌絲生長及多醣之生成均有促進作用。因此,研究所得之較佳培養基組成為20 g/L brown sugar 、10 g/L malt extract、5 g/L yeast extract、5 g/L skim milk、2 .5g/L紅花籽油、0.5g/L CaCo(下標3)、0.5 g/L K(下標2)HPO(下標4)、0.5g/L KH(下標2)PO(下標4)和0.5 g/L MgSO(下標4).7H(下標2)O。此發酵系統較佳之起始 pH 值及培養溫度分別為 6.5和 34℃,搖瓶轉速則以高轉速(160 rpm)為宜,以提高氧氣傳送速率。依上述條件進行培養時,所得菌絲體產量(7.25 g/L與多醣體生成量(0.265 g/L)皆較探討前有大幅提升。 |
