Description

Abstract
"霍亂弧菌常發現在一般水環境及水產動植物之體表,可依據O-抗原分類,目前已有超過200種以上之血清型。但僅有O-1及O-139兩種血清型會有霍亂毒素,並引起霍亂。致病性之霍亂弧菌是因為含有霍亂毒素基因(ctx基因)。而此兩種血清型之霍亂弧菌比其他霍亂弧菌不易被分離纯化,因為霍亂弧菌有時會呈休眠狀態。致病性之霍亂弧菌雖呈休眠狀態,但具有爆發霍亂之潛力,因此快速檢測致病性霍亂弧菌之方法極為迫切需要。本計畫主要利用聚合酶鏈反應及rfb基因引子與ctx基因引子區分霍亂弧菌及是否具有毒性,以建立一個有效的系統可提供資料及分析致病性霍亂弧菌爆發之潛力。" "Vibrio cholerae is a free-living bacterium found in many aquatic environments and in association with plankton, plants, invertebrates, and fish. Vibrio cholerae is a heterogenous species, with more than 200 serotypes identified to date, based on the heat-stable somatic O antigen. Only two serotypes, V. cholerae O1 and O139, have been characterized as toxigenic and recognized as the causative agent of epidemic cholera; however, they are less frequently isolated in aquatic environment than non-O1/non-O139 strains. Both pathogenic serogroups contain ctx gene related to pathogenicity. Vibrio cholerae is native to brackish and estuarine environments and is associated with zooplankton, mainly copepods. Detection of V. cholerae may be difficult because this microorganism can be found in two forms: viable and culturable and viable but non-culturable (dormant state). Because of the devastating epidemic potential of O1 and O139 serogroups, the quick diagnosis and identification of the the causative serogroup is important especially from a public health point of view. For surveying Vibrio cholerae in aquatic enviroments, total DNA extraction, followed by V. cholerae PCR, and O1/O139 serogroup and ctx genes by PCR offers an efficient system, providing information on the toxigenic potential of the strains and permitting risk analysis for cholera in aquaculture."
