Description

Abstract
"石斑魚因鮮美的肉質及生長快速等的優勢,成為台灣養殖產業上重要的經濟魚種,但隨著產業的發展病毒的爆發將成為發展的阻礙。目前雖有商用檢測試劑,但其靈敏度、方便操作卻仍有改善的空間,故研發一套快速、靈敏的疾病檢測平台是現今迫切需要的。磁量生技公司的專利MagQu免疫磁減量檢測系統,是利用標誌抗體的磁珠與病毒結合後產生的磁減量現象,將微弱的磁訊號轉變為可量測的電壓訊息,具相當高的靈敏度。本實驗室過往與磁量生技合作上,已開發一套快速靈敏且不需專業人士操作的病毒檢測平台,並在去年度農業生物技術國家型科技計畫中建立了一個病毒感染及發病的資料庫,從這個資料庫可提供第一線養殖業者不僅檢測養殖池中的病毒量,還可做為是否繼續畜養的根據以減少損失。為了使檢測套組的準確性更高且能夠提供品質穩定的抗體,本計畫利用單株抗體具有對特定抗原有高度專一性的特性,製備單株抗體標定於磁珠上,除了台灣本土的魚類病毒之外,本計畫亦考慮到未來國際的競爭力,同時篩選國外的病毒株製作單株抗體,此技術不僅可促進檢測套組的效能,還有助於其他包括抗病製劑等生技產業之發展。" "Grouper has becomes economically high-valued aquaculture species in Taiwan due to advantages such as great quality of flesh and fast growing rate. However, the outbreaks of viral diseases become hindrance to the development of this industry. Although the commercial detection test kit is available, there are rooms for improvements in sensitivity, easiness of operation and etc. A magnetic immunoassay analyzer which is patented by MagQu Co., Ltd., can detects the magnetoreduction of virus and the magnetic beads coated with targeted antigen, and further convert this immunomagnetic reduction (IMR) to the measurable alternating current reading. In previous the cooperation of MagQu Co., Ltd. with our laboratory, we established a systematic viral detection platform and database for viral infection and outbreak in National Science and Technology Program for Agricultural Biotechnology. This database not only can provide the farmers with the viral quantity, but also provide the foundation in determination of continuing cultivation to prevent or reduce lost. In order to increase the accuracy of the detection kit and a stable quality of antibody, our project proposes the monoclonal antibody production for the magnetic bead coating as it has the higher specificity to desired antigen. As the consideration of international competitiveness, this project selected both Taiwan’s and oversea virus for the monoclonal antibody production. This monoclonal antibody production eventually can increase the efficacy of the detection kit and can also promote the development in biotechnological industry."
