Description

Abstract
"本研究針對兩種石斑魚虹彩病毒病毒,開發複合式口服雙價病毒DNA疫苗。一劑中包含兩種病毒之次單位疫苗,且可以添加飼料,經口投餵魚苗,符合養殖產業需求。首先個別選殖兩種虹彩病毒構造蛋白中,最具免疫原性之主要外鞘蛋白基因,並選殖入真核用表現載體,經確認所建構載體表現無誤後,轉殖入細菌並大量培養,萃取蛋白質表現載體即為DNA疫苗。分別製作不同病毒DNA疫苗,之後進行兩階段乳化包埋處理,所得到之最終產物即為口服多價病毒DNA疫苗。同時在這個計畫當中,也會評估市售的各類佐劑及中草藥來做為DNA疫苗的佐劑,增強DNA疫苗的效果並降低未來市售的成本,除此之外,包含DNA疫苗及佐劑的免疫機制也是本計畫想要釐清的重點,在這個計畫當中,包含:1、疫苗的傳遞方式;2、疫苗佐劑作用機制;3、疫苗效力測試。這三方面將會跟大陸中科院南海所秦啟偉、水生所張奇亞研究員做緊密的合作。" "In our study, an oral bivalent DNA vaccine against two type iridovirus of grouper in Taiwan are developed. Single serving contains two types of DNA vaccine and can be administered orally with feed, will be an idea vaccine product for aquaculture industry. First, virus major capsid protein gene with best immunogenicity are cloned and ligates into a prokaryotic expression vector respectively. These constructed vectors are then transformed into bacteria and applies to massive culture bacteria. Afterwards, extract recombinant protein expression vector (as DNA vaccine). Subunit vaccine of different viruses is individually produced and used as the first liquid. The first liquid mix with second mixture (edible oil) to produce the first emulsions. Subsequently, mix all of the above emulsions equivalently into a third solution. And the final emulsions obtained from the above two-step multiple emulsified techniques is an oral bivalent DNA vaccine product. At the same time, we will also evaluate the commercial adjuvants and Chinese herbs as DNA vaccine adjuvants to improve the vaccine efficiency and reduce the cost of the DNA vaccine. On the other hand, we will also investigate the immune mechanism of DNA vaccine and adjuvants. We will also tightly collaborate with Chinese researchers; Dr. Qi-Wei Qin and Qi-Ya Zhang with following aspects. 1. Vaccine delivery 2. Mechanisms of DNA vaccine and adjuvants 3. The efficacy of DNA vaccine."
