Abstract
"水產養殖乃人類食物主要生產來源,而吳郭魚為世界主要養殖魚類物種之一,在提供食用優質蛋白方面扮演著至關重要的角色。然而,疾病爆發近來已成為吳郭魚養殖的主要瓶頸,並時而導致巨大損失。了解這些養殖生物之各免疫相關基因、細胞及其反應、機制和功能,乃為疫苗設計、抗病品系選擇和預防疾病爆發之首要關鍵。先天免疫系統在宿主防禦病原體中扮演著關鍵角色,其使用胚系基因”模式識別受體”識別由入侵病原體帶有之特定、保守且對於微生物的存活至關重要、難以任意改變的組成產物,又稱為病原體相關分子模式。本計劃將著重研究吳郭魚類鐸受器 (TLR) 18之免疫功能。我們將搜索基因組、選殖及定序類鐸受器18及類鐸受器1亞家族之成員,並將通過生物信息學剖析其功能結構域之組成、基因同源性和與基因共線性區塊。此外,我們將偵測此些基因在體外和體內不同的刺激或感染模式下表現量之調節,以幫助了解這些基因在發炎期間或針對病原體的免疫應答中之作用與相關資訊。我們亦將在魚組織和表達質粒轉染的細胞中研究該受器於細胞表現之位置。最後,我們將開發細胞系平台及類鐸受器18基因剔除魚品系,以研究其信號傳導途徑和配體特異性。" "Aquaculture is known as an important food-producing industry and tilapia is the major cultured species worldwide which plays a vital role on providing high quality protein for human consumption. However, infectious disease outbreaks in farmed tilapia have become an emerging problem recently and commonly resulted in a tremendous cost to the industry. Hence, understanding what immune-related genes and cells are present, their responses, mechanisms and functions in these farmed animals is a first requirement for potent vaccine design, selection of disease-resistant breeds and disease outbreak prevention. The innate immune system plays a key role in the defense of the host against pathogens which use germline-encoded pattern recognition receptors (PRRs) to recognize specific, conserved and constitutive products of invading pathogens, called pathogen-associated molecular patterns (PAMPs) that are important for survival of the microorganism and are thus hard for the microorganism to change. This project focuses on studying the immune function of TLR18 in Nile tilapia (Oreochromis niloticus). TLR18, and other members in the TLR1 subfamily will be identified, cloned and sequenced and their putative domain structure, gene synteny and homology to other genes will be determined by bioinformatics analysis. In addition, the potential modulation of these genes will be examined in different in vitro and in vivo models which provide information to help understand the role(s) of these genes during inflammation or in the immune responses against pathogens. The sub-cellular localization will also be investigated in fish tissues and expression plasmid transfected cells. Finally, we will develop cell line based platforms as well as TLR18 knockout fish to study TLR18 signalling pathways and ligand specificity."
