Abstract
作為『副熱帶陸棚浮游生物食物網生態過程研究』整合研究計畫之子計畫之一,本計畫主要想了解寡毛亞綱纖毛蟲在東海南部的種類組成時空分布。寡毛亞綱纖毛蟲扮演著將有機碳能量由微生物食物網傳遞至高階傳統食物鏈的重要角色。由於寡毛亞綱纖毛蟲歧異度極高,同時其營養模式非常多樣,因此在討論碳循環或環境變遷時,研究寡毛亞綱纖毛蟲的群聚結構就成為一個重要且必要的課題。為了探討研究海域寡毛亞綱纖毛蟲數量變化及種類組成,本研究不僅於倒立式顯微鏡下觀察計數寡毛亞綱纖毛蟲,同時以直徑47 mm大小的polycarbonate濾膜(Millipore)進行海水過濾以獲得寡毛亞綱纖毛蟲18S rDNA V4段序列。以序列作為種類之依據,再搭配顯微鏡計數的生物量,不僅能進一步了解東海南部海域寡毛亞綱纖毛蟲種類多樣性,同時也可探討其族群結構隨著空間與時間的變化過程。對東海南部馬祖至黑潮(小東海)測線之小尺度生態研究將有顯著的貢獻。最後所得之序列資料將作為東海南部海域寡毛亞綱纖毛蟲種類組成之資料庫,增加後續研究之背景資料能量。目前研究結果發現同一航次因環境的不同而有不同的類群組成及豐度變化;而同一採樣測站不同月份所採集的樣本,其種類組成及豐度亦有不同的表現。此研究數據在在顯示出纖毛蟲的類群組成及豐度對環境反應迅速。The aim of this project, a component of the cooperative research program entitled "Plankton Ecological Processes in Subtropical-shelf Ecosystems" (PEPSE), is to understand the spatial and temporal distribution and composition of oligotrich ciliates in the south of the East China Sea. Oligotrich ciliates play an important role in the trophic link from the microbial food web to the traditional food web in the open ocean. With the highly biodiversity and variable trophic strategies in oligotrich ciliates, understanding its community structures when discussing the carbon cycles and/or environmental changes is an essential work. In order to discuss the variation of the abundance and the community composition of the oligotrich ciliates, not only will oligotrich ciliates be counted under an inverted microscope but also the seawater samples will be filtered onto a 47 mm polycarbonate filter membrane (Millipore) for extracting V4 regions of 18S rDNA sequences. With the data of sequences as species and their abundances, we can better understand the diversity and community structure under the spatial and temporal scale. A database of the composition of oligotrich ciliates will be made up by the V4 regions of the 18S rDNA sequences, for the background data of the research in the future. According to the preliminary data, we found variety of oligotrich ciliate communities and abundances in each sampling site of the same cruise and in the same sampling site of different cruises. All the data indicate the rapid response of the diversity and abundance of oligotrichs to the shift of the environment.
