Description

Abstract
觀賞魚的經濟價值一般決定於魚體體型、體表顏色、體表變化多樣的圖紋以及近年來逐漸受到消費者重視。其中體色的鮮豔度及體表圖紋的花樣更是業界育種改良的重點。血鸚鵡、花羅漢、麒麟鸚鵡等慈鯛(麗魚屬)魚種為台灣自行開發之重要觀賞新品種。魚類體表色素與圖案之生成,色素細胞與色素之分佈均與基因表現與調控有關。本計畫之目標是麗魚屬(Cichlasoma)血鸚鵡為模式動物魚種來研究觀賞魚的色素 (pigmentation)與圖案(pattern)生成之相關基因與調控機制,並研究各種生理因子對體表色素與圖案調控相關基因表現之影響。本計畫亦將本計畫結果應用於血鸚鵡揚色、增豔飼料上。本年度研究目標:1)將以分子生物技術選殖血鸚鵡色素調控因子酪胺酸酶與觀賞魚身體圖案形成之調控因子Mitf與PSTS等基因與特性分析;2)依據麗魚屬各魚種色素與圖案生成基因啟動子之特性,研發促進色素與身體圖案基因啟動子之化學藥物、神經傳導物質、性賀爾蒙,中草藥等促進血鸚鵡之體表紅色素形成與沈積;本計畫之效益是多層的:1.增加對色素基因在低等脊椎動物表現之調控機制了解,進而操控基因表現2.研發促進血鸚鵡色素與身體圖案基因啟動子之化學藥物、神經傳導物質、性賀爾蒙、中草藥等做為飼料添加劑,以促進血鸚鵡體表紅色素生成與沈積,達到觀賞魚揚色、增豔之效果,使我國觀賞魚產業更具國際競爭力,增加觀賞魚之附加價值,提高利潤。 Ornamental fish production for the aquarium industry is a multibillion dollar industry in the Asia countries. In ornamental high-value species such as cichlasoma, and goldfish (Carassius auratus), emphasis should be given to achieving high levels of skin pigmentation, which, together with body shape, fin shape and body size, are the most important quality criteria informing their market value. The colours and patterns of the skin provides a fascinating system used for the study of pattern formation in experimental and theoretical research alike. Since 1945, cichlids have become increasingly popular as aquarium fish Cichlids are ideally suited as aquarium fish.Cichlasoma is a genus of fish in the cichlid family. The most ubiquitous aquarium hybrid is perhaps the blood parrot cichlid which is a cross of several species especially those from genus cichlasoma. There are several gene requirements in vertebrate melanocyte development and regulate pigment pattern. Fish of the family Cichlidae are particularly known for brilliant coloration especially in mature males. Therefore, cichlasoma is suitable as animal model to study genetic pigment pattern. In the present study, we propose using cichlasoma species as model fish to study genetic pigmentation and pattern. To fulfill this objective, we will establish following short term studies; 1) molecular cloning and characterization of tyrosinase gene and pigment pattern regulator genes, such as Mitf and PSTS genes. 2) to determine sex steroids, neurotansmitters influence pigementation in blood parrot cichlid. 3) to employ these results to develop commercial color enhancing diet for blood red parrot . We will expect that the outcome of this project will be several-fold, First, to elucidate regulator genes action pigmentation and pattern on cichlasoma. Second, to develop a high effectively color enhancing diet for commercial ornamental fish purpose. The application of biotechnology in ornamental fish could provide a significant competitive edge for Taiwan ornamental fish industry wise enough to explore its benefits.
