Description

Abstract
我們先前研究已證實抗菌肽可在細胞膜上造成暫時性 ”孔洞(pore)”。是一種廣譜性殺(抑)菌。其中有一個抗菌肽-表面素,surfactin既為抗菌肽,又為生物介面活性劑biosurfactant,先前試驗結果表明surfactin是一種乳化劑(emulsifier)與皮膚助滲劑(transdermal penetration enhancer)。surfactin可造成在水溶液途徑中延長增加分子的透皮累積。在本計畫目標是將水產生物抗菌肽應用於化妝保養品。為達成此一目標我們設立下列短程目標;一)篩選高量產surfactint突變株,並建立實驗室級試量產規模生產技術;二)建立優化生產條件與純化方式;三)開發此類新型抗菌肽在化妝保養品之天然防腐劑之應用;四)開發此類新型抗菌肽作為化妝保養品之天然乳化劑;五)開發此類新型抗菌肽作為化妝保養品之皮膚助滲劑(TPE)。將此類創新天然抗菌肽助滲劑應用在化妝保養品的大分子滲入皮膚深層,也可作為加強各種劑型,包括貼布、藥膏等醫療藥物滲入皮膚。本創新型抗菌肽可作為天然防腐劑、乳化劑與皮膚助滲劑,其應用層面廣泛,包括:食品業化妝保養品、醫療、製藥業),具有巨大商機。 The present proposal relates to novel antimicrobial peptides (AMPs), from tiger shrimp (Penaeus monodon; monodoncin), grouper and Bacillus sp. These AMPs can be overexpressed in a heterologous expression system, such as yeast, by molecular cloning techniques or by induction technique. It had been demonstrated a wide-range of bacteriostatic and bactericidal effects on gram (-) and gram (+) bacteria, as well as fungicidal activities. This novel natural antimicrobial peptide can be used as preservative of cosmetics. We had isolated a novel surfactin (C53 H 93N7O13) is a biosurfactant which is produced through fermentation and purification of high-yield mutant Bacillus strain. The results of our previous studies demonstrated that this novel surfactin not only has anti-bacterial, anti-fungal and anti-virus effects, but also having emulsification and trnasdermal penetration enhancer activities. The goal of present proposal is application of aquatic antimicrobial peptides in cosmeceutic products. To fulfill this objective, we have established following short-term goals; 1) to screen high yield surfactin strain; 2) to optimize producting surfactin fermentation and purfiction conditions for surfactin yield, 3) to employ of aquatic AMP to suppress microbes, reaching has pathogens free states, as a natural preservative additive in the cosmetics and skin care products.; Promotion the emulsification of the oil and water, surfactant is applied . In the past most surfactants are chemical surfactants, harmful to often have poisonning or its supersession derivative to the environmental ecology as well. Therefore, Natural surfactants produced from microorganisms, plants or animals are referred to as biosurfactants which are biodegradable, friendly to the environmental ecology is needed; In present proposal, 4) to employ surfactin as an emulsifier in in cosmeceutic products. 5) to determine the application of surfactin as a transdermal penetration enhancer for enhancing large molecules to penetrate skin layers.
