Description

Abstract
計畫目標: 與魚類成長有關之重要內分泌因子為生長激素與類胰島素生長因子, 而生長激素主要為促進肝臟分泌類胰島素生長因子, 進而促進魚類之成長作用, 因此生長激素應是促進魚類成長最重要之激素, 如何調控魚類生長激素的合成與分泌, 應是養殖上重要的課題.因此本研究目標擬建立一套評估各種活性物質促進生長激素之分泌, 並瞭解生長激素之分泌機制.架構: 1.建立黑鯛腦下垂體細胞初級培養之技術 2.測驗各種可能之內分泌激素或活性物質對腦下垂體細胞分泌生長激素之作用.預期效益: 將可建立黑鯛腦下垂體細胞初級培養之系統, 並可供為評估各種物質調控腦下垂體細胞分泌生長激素之作用, 進而開發有用之活性物質. Growth hormone and insulin-like growth factor are the important endocrine factors involving in the growth-promoting effects.Growth hormone mediated the growth performance in fish through the mediation of insulin-like growth factor.The regulation of growth hormone release is an important aspect for the control of fish growth.There are many endocrine factors or synthesized compounds may have the potency to regulate the release of growth hormone.Therefore, the objectives of the study are to develop an in vitro system to evaluate the stimulation of growth hormone release in black porgy by applying several compounds.Pituitary cells of black porgy will be dispersed and a static culture of the pituitary ceels will be developed.Several endocrine hormones and synthetic compounds will be applied to the culture medium in the static culture system.Growth hormone concentrations are measured by a specific radioimmunoassay.By conducting this experiment, we expect to accomplish the development of a static culture system of the pituitary cells to further evaluate the possible growth-promoting compounds.The regulatory system of growth hormone will also be understood.
