Description

Abstract
在胚胎發育過程中,視黃酸透過細胞的視黃酸接受器訊息傳遞系統調控下游目標基因的活性。在哺乳動物與魚類的胚胎發育過程中,過量的視黃酸會改變細胞內各重要基因的活性進而影響到胚胎顱面、心臟、四肢或胸鰭、中樞神經以及其他器官的發育。這些參與細胞分化與胚胎發育的基因調控子除了各種的Homeoproteins等DNA辨識蛋白之外,尚有許多其他的DNA辨識蛋白亦扮演了相當重要的角色,例如bHLH-PAS蛋白在魚類心臟、胸鰭及腦的發育扮演相當重要的功能,但其分子機制則尚不清楚。本計畫即是針對最近由本實驗室所選殖出的一系列斑馬魚bHLH-PAS基因來探討對胚胎發育所扮演的角色,並且了解視黃酸對這些基因的影響。 Retinoids are the metabolites of vitamin A which play important roles in embryo development. Excess or deficiency of retinoic acid causes abnormal development in vertebrate heart, cranial and central nervous system. The exogenous retinoic acid disturbed the genetic activity of a number of vertebrate homeotic genes during embryogenesis. In addition to the homeoproteins, a novel bHLH-PAS protein family were also revealed to act important roles in vertebrate development. Members of bHLH-PAS proteins include aryl hydrocarbon receptor (AHR) and its associate factor (ARNT), SIM, PER, CLOCK, and hypoxia-inducible factor, HIFs. In previous study, we have identified a group of bHLH-PAS factors from zebrafish and they all exhibit specific expression pattern in the developing embryos. In this study, we have characterized the spatiotemporal expression pattern of SIM2 and HIF 1alpha, HIF 3alpha genes in zebrafish embryo. The effect of retioic acid in these gene expression were evaluated by in situ hybridization.
