Description

Abstract
本計畫於第一年度主要工作在探討低氧誘發蛋白HIF-1α與HIF-3α對於頭骨與顱面的發育影響。首先利用軟骨染色的方法觀察HIF-1α與HIF-3α基因knockdown對胚胎頭骨的影響,結果顯示軟骨細胞排列紊亂,無法成型。隨後利用標識基因追蹤各胚層的發育情況,發現HIF-1α與HIF-3α基因knockdown會造成內中胚層均受到嚴重的影響,造成內胚層所衍生的咽弧,咽囊構造,以及中胚層所發育的顏面肌肉等均有明顯的缺損現象,而頭骨的先驅神經脊細胞的遷移亦受到影響,以致於軟骨的構造排列凌亂,並且大量減少的現象。此外,菱腦的分節亦有緊縮的現象。我們的實驗結果,顯示缺氧細胞訊息對於正常胚胎發育扮演有相當重要的功能。這項工作將持續追蹤較早期的胚胎發育狀況,以及探討腦部重要器官如下視丘,腦下垂體等的影響。 The function of Hypoxia-Inducible Factors, HIF-1a and HIF-3a, in head skeleton and cranial facial development was investigated. Gene-specific morpholino oligonucleotide was injected into fertilized zebrafish embryos at 1-cell stage to observe the effect of gene-knockdown in embryonic development. It appeared that the craniofacial development was disrupted in hif-1a & hif-3a-knockdown embryo. A number of endodermic and mesodermic markers were also blocked in these embryos. Consequently, the pharyngeal pouches and cranial muscle was defected in hif-1a & hif-3a-knockdown embryo. In addition, the neural crest cells migration and segmentation of rhombomere was disturbed by hif-1a & hif-3a morpholino oligonucleotides. These observations suggest that the HIF-1a and HIF-3a-related signaling pathways have important roles in vertebrate development. We will continue to work on this project and investigate the function of HIF-1a and HIF-3a-related signaling in brain development.
