Description

Abstract
"本計晝之目標是合成功能性奈米複合材料並結合雷射脫附游離質譜或仿生酵素特 性於腫瘤組織顯影。首先會先合成氧化石墨烯或金屬氧(硫或滷)化物奈米片載體,再將 辨識分子(適合體和蛋白質等)修飾的金屬(金或銀等)奈米粒子載上氧化石墨烯或金屬 氧(硫或滷)化物奈米片。在雷射脫附游離質譜系統中,可觀察複合材料選擇性標示於腫 瘤組織後,於脈衝雷射擊打後所解離的金屬團蔟離子的訊號來進行組織顯影。以仿生 催化系統中,則是藉由奈米複合材料選擇性標示腫瘤組織後,其特殊仿生催化特性來 達螢光或顯色的組織顯影效果。我們所開發的高靈敏度、高解析度與快速組織顯影技 術未來將可應用於腫瘤細胞的快速辨識與臨床初期癌症診斷。 " "The goal of this proposal is to combine nanocomposite materials with laser desorption/ionization (LDI)-mass spectrometry or its unique enzymatic catalysis property for the imaging of tumor tissues. First, we will focus on the synthesis of nano-supporters of graphene oxide (GO) and metallic oxide (sulfide or halide) nanosheets. After that, aptmer- or antibody-modified metal nanoparticles (e.g., gold nanoparticles, silver nanoparticles) will be loaded on the nanosheets, and the resulting nanocomposites will be used for the specific labelling of tumor tissues. The imaging of the tumor tissues can be achieved by observing the metallic cluster ions produced when the nanocomposites-labeled tissues are irradiated with pulsed laser during the LDI mass spectrometry. Another strategy for imaging of tumor tissues in enzymatic catalysis system is by fluorescence or absorption spectroscopy method. Our preliminary experiments on this proposal suggest that these two detecting systems not only can be employed for the analysis of tumor cells in tissue but also for the early diagnosis of cancer. "
