Abstract
本計畫將以重組DNA技術,將四種具有抑制ACE活性之機能性胜肽(AW、IVY、LIY、LKPNM)的相對DNA序列經適當排列組合後,插入適當之蛋白質表現質體,於大腸桿菌中大量表現含有重組多胜肽的融合蛋白,純化的重組多胜肽由可再經由chymotrypsin作用於W, Y及M殘基之脧基端胜肽鍵,可產生含有四種可抑制ACE活性之機能性胜肽的混合物.本計畫之預期效益為 (1) 建立新型之機能性胜肽生產技術,以因應未來巿場對於機能性胜肽的需求. (2) 由大腸桿菌生產重組多胜肽之方式建立後,未來可再將此系統修飾後轉入麵包酵母菌,直接由GRAS微生物來產生機能性胜肽. This study aims to produce four functional peptides (AW, IVY, LIY, and LKPNM), which can inhibit the activity of angiotensin I-converting enzyme, by recombinant DNA technology. After a proper arrangement of the corresponding DNA sequence of these four peptides, the DNA sequence will be inserted into a protein expression vector of E. coli. This fusion protein will be overexpressed in E. coli. After purification chymotrypsin will be used to digest the polypeptide by cleaving the carboxylic ends of the residues, W, Y, and M. The digested mixture will contain multiple copies of those four functional peptides, which might have synergistically inhibitory effect on the angiotensin I-converting enzyme. This study is expected to establish a technology for the production of functional peptide to meet the future demand and to use a GRAS host, such as baker's yeast, in the peptide production after the establishment of the production in E. coli.
