Description

Abstract
為了解膳食幾丁聚醣對脂質代謝與免疫功能的影響,以雄性Wistar或SD(Sprague Dawley)大白鼠為實驗動物,分別進行以下實驗。實驗(一):餵食Wistar大白鼠5%纖維質(控制組),或5%幾丁質或5%幾丁聚醣或7%幾丁聚醣8週後,發現幾丁聚醣組有明顯較低的血漿IgA濃度,但是IgG濃度方面四組間在統計上無明顯差異。大白鼠攝食幾丁聚醣後有較高的糞便總脂質及較低的肝臟總膽固醇濃度。此外幾丁聚醣也會降低肝臟葡萄糖六磷酸去氫?、葡萄糖六磷酸?及六碳醣激?等酵素活性。實驗(二):以SD大白鼠為實驗動物,分別餵食5%纖維質(控制組) ,幾丁聚醣(以化學法生產)或幾丁聚醣(以微生物分解生產)等四週,結果發現不論是以化學方法生產或是微生物生產的幾丁聚醣都會明顯下降血漿總膽固醇,LDL膽固醇及VLDL膽固醇濃度。雖然三組間在體重上沒有明顯差異,但是攝食幾丁聚醣後有較低的肝體比。比較化學法或微生物生產的幾丁聚醣對大白鼠脂質代謝的影響發現,大白鼠攝食化學法所生產的幾丁聚醣後有較低的肝臟總脂質、總膽固醇及較高的肝臟脂質過氧化物濃度。此外,分析兩種幾丁聚醣物化性質發現,化學法生產出來的幾丁聚醣有較高的黏度與分子量。綜合以上結果,推測(1)長期攝食幾丁聚醣可能會下降免疫能力,(2)膳食幾丁聚醣可能會下降肝臟脂質合成酵素及糖類代謝酵素,(3)不同方法所生產出來的幾丁聚醣可能對大白鼠肝臟脂質代謝的影響有所不同,其最大的原因可能與幾丁聚醣的分子量或黏度的不同有關。 To investigate the dietary chitosan on lipid metabolism and immunity function, male Wistar or SD (Sprague Dawley) rats were used as animal model. Exp. (I): Wistar rats fed 5% chitosan for 8 weeks have lower plasma IgA concentration when compared with those fed cellulose diet. No significant change in plasma IgG concentration was found among the dietary groups. Chitosan significant decreased liver total cholesterol and increased fecal total lipids. In addition, significantly decreased liver glucose-6-phosphate dehydrogenase, hexokinase and glucose-6-phosphate activities were observed in rats fed 5% chitosan. Exp (II): SD rats were fed a diet containing 5% cellulose, or CCS (chitosan manufactured by chemical method), or MCS (chitosan manufactured by fermentative method) for 4 weeks. A significant decrease in plasma total cholesterol, LDL-cholesterol and VLDL-cholesterol were found in rats fed a diet containing chitosan. Although no significant difference in body weight was found among the dietary groups, rats fed the chitosan diet have lower ratios of liver weight to 100g B.W. when compared with those fed cellulose diet. Lower liver total lipids and total cholesterol contents and higher liver peroxide concentration were observed in CCS group when compared with MCS group. Lower intrinsic viscosity and molecular weight were found in chitosan manufactured by chemical method than chitosan manufactured by fermentative method Results from this study suggest that (1) dietary chitosan may play an important role in regulation of lipid metabolism and immunity function in rats. (2) the different physiologic effect of the two chitosan in rats may be related to the different viscosity and intrinsic viscosity properties.
