Description

Abstract
為了解幾丁聚醣在添加聚醣後對於正常及糖尿病大白鼠血脂質、血糖及腸道生理之影響,以48隻13週雄性SD大白鼠為實驗動物,分成四組分別為:(1)Cellulose non-diabetic group;(2)Chitosan non-diabetic group;(3)Chitosan+Inulin non-diabetic group;(4)Cellulose diabetic group;(5)Chitosan diabetic group;(6)Chitosan+Inulin diabetic group;實驗共進行7週,實驗期間採自由飲水攝食的方式。實驗結果發現,在正常大白鼠方面幾丁聚醣組及幾丁聚醣添加菊醣組會明顯有較低的體重及脂肪組織重量;而在糖尿病大白鼠方面,幾丁聚醣組及幾丁聚醣添加菊醣組則會有較高的體重,但在脂肪組織重量上並無明顯差異。血脂質的變化方面,幾丁聚醣及幾丁聚醣添加菊醣皆會顯著性的降低正常及糖尿病大白鼠血清總膽固醇濃度。值得注意的是,幾丁聚醣及幾丁聚醣添加菊醣組之糖尿病大白鼠血清膽固醇濃度下降之程度分別高達81%及75%,而血清TBARS濃度也明顯降低,而以幾丁聚醣添加菊醣組最低。在肝臟脂質方面,無論在正常及糖尿病大白鼠方面,幾丁聚醣及幾丁聚醣添加菊醣組皆會明顯降低肝臟三酸甘油酯及膽固醇含量,並提升肝臟TBARS濃度。而在糖尿病大白鼠方面,幾丁聚醣添加菊醣則較幾丁聚醣組有較高的三酸甘油酯與膽固醇含量,以及較低的肝臟TBARS濃度。在血糖濃度方面,幾丁聚醣及幾丁聚醣添加菊醣組並不影響正常大白鼠血糖的濃度,但卻會分別降低糖尿病大白鼠血糖濃度達49%及46%。此外,無論正常或糖尿病大白鼠,攝食幾丁聚醣及幾丁聚醣添加菊醣後皆會增加盲腸重量及小腸長度。有趣的是,攝食幾丁聚醣會提升正常大白鼠盲腸Ph值,但此種現象並未表現在糖尿病大白鼠上,而無論正常或糖尿病大白鼠,攝食幾丁聚醣添加菊醣後皆會明顯降低盲腸的pH值。綜合以上結果得知幾丁聚醣不論在正常或糖尿病大白鼠皆有降低血膽固醇的功能,而對於糖尿病大白鼠而言,幾丁聚醣不但有助於脂質代謝,對糖代謝也有很大的改善。而菊醣的添加則有助於降低腸道pH值,並減少糖尿病大白鼠體內脂質的過氧化作用。 To investigate the effect of chitosan on plasma lipids and intestinal physiology in normal and diabetic rats with inulin supplementation, male Sprague Dawley (SD) rats were used as experimental animals and divided into six groups. There are (1) Cellulose group (CE) (2) Chitosan group (CS) (3) Chitosan with inulin supplementation group (CSI) (4) Diabetic cellulose group (DCE) (5) Diabetic chitosan group (DCS) (6) Diabetic Chitosan with inulin supplementation group (DCSI). The experiment was done for 7 weeks. Significantly decreased plasma total cholesterol, VLDL-cholesterol and LDL-cholesterol were observed in normal and diabetic rats fed a chitosan diet. In addition, chitosansignificantly decreased plasma glucose and free fatty acidlevels in diabetic rats. Although chitosan significantly increased cecum pH value, lower cecum pH value was observed in normal rats fed a chitosan diet after inulin supplementation. Higher lipid peroxide value was observed in rats fed the CS diet or DCS diet when compared with those fed the CE diet. However, inulin treatment significantly decreased the liver lipid peroxide value. Results from this study suggest that chitosan may affect the plasma lipids, liver lipids, liver peroxidation and cecum pH value in normal and diabetic rats, and inulin supplementat may regulate the lipid metabolism and in testine physiology in rats fed a diet containing chitosan.
